摘  要:补骨脂素和异补骨脂素是中药补骨脂的特征成分,含量较高且具有多种生物活性,目前已成为补骨脂及其产品的质量控制成分。本文以补骨脂干燥成熟果实为原料,经乙醇回流制备补骨脂粗提物,率先采用高速逆流色谱技术进行分离,所得样品由高效液相色谱检测纯度,并由对照品确定成分。实验结果表明:采用正己烷-乙酸乙酯-甲醇-水(7:3:5:5)作为两相溶剂体系,经一步逆流分离可以从补骨脂粗提物中分离得到补骨脂素和异补骨脂素,二者纯度分别达到99.4%、99.1%。该法具有制备量大、分离效率高等优点。High-speed counter-current chromatography (HSCCC) was employed for the separation of psoralen and isopsoralen from a crude extract of Psoralea corylifolia. The crude extract was obtained by extracting with ethanol from the dried seeds ofPsoralea corylifolia, n-Hexane-ethyl acetate-methanol-water (7 : 3 : 5 : 5)was used as the HSCCC two-phase solvent system for the separation. The isolated high-pure components were analyzed by high performance liquid chro- matography (HPLC) and identified by comparing of the retention time with those of the standards. The results indicated that psoralen and isopsoralen were separated from other elements at 99.4% and 99.1% purity. High-speed counter-current chromatography could separate and prepare these two components successfully from Psoralea corylifolia seeds.
