摘 要:【目的】观察黄芪注射液对腹膜透析大鼠腹膜间皮细胞水孔蛋白1(AQP-1)表达的影响。【方法】选用雄性SD大鼠25只,分正常对照组(N=7)、模型组(N=9)和黄芪组(N=9)。除正常对照组外,其他两组分别腹腔注射含42.5g/L葡萄糖的腹透液和含42.5 g/L葡萄糖的腹透液+黄芪注射液(20 mg/mL),剂量为25 mL/d,连续10 d。于第11天观察大鼠透析超滤量(UF)与净超滤量(NetUF)、腹膜钠转运(D/P Na)、腹膜组织光镜病理形态及AQP-1免疫组化的表达。【结果】透析液留腹60 m in时黄芪组UF、NetUF与模型组比较显著增加(P<0.05)。透析液留腹30 m in、60 m in时黄芪组D/P Na较模型组显著减低(P<0.05)。黄芪组大鼠腹膜层增厚、间皮细胞脱落情况较模型组减轻,AQP-1在腹膜间皮细胞的表达较模型组显著增多(P<0.05)。【结论】黄芪注射液可减轻高浓度葡萄糖腹透液对间皮细胞的损伤,升高AQP-l在腹膜间皮细胞的表达,改善透析液留腹早期跨细胞水转运功能,从而提高腹膜对水的清除,增加透析超滤量。Objective To observe the effect of Astragalus Injection (AI) on aquaporin-1 (AQP-1) expression of peritoneal mesothelial cells in rats with peritoneal dialysis. Methods Twenty-five male SD rats were divided into the normal control group ( N = 7 ), model group ( N = 9 ) and AI group ( N = 9 ). Except the normal control group, the model group was given intraperitoneal injection of peritoneal dialysate ( containing glucose 42.5 g/L), and AI group was given intraperitoneal injection of peritoneal dialysate (containing glucose 42.5 g/L) plus AI (20 mg/mL), 25mL/d for 10 continuous days. On the 11^th day of experiment, the amount of dialysis ultrafiltration (UF) and net UF, and the concentration ratio of dialysate and plasma sodium (D/PNa) were measured, peritoneal pathological changes were observed with light microscope, and the expression of AQP-1 in peritoneal mesothelial ceils was detected with immunohistochemical method. Results When the peritoneal dialysate was kept in the abdominal cavity for 60 rain, the amount of UF and Net UF was increased in AI group ( P 〈 0. 05 compared with that in the model group). When the peritoneal dialysate was kept in the abdominal cavity for 30 min and 60 rain, D/P Na in AI group was lower than that in the model group (P 〈 0. 05 ). In comparison with the model group, the changes of thickened peritoneal layer and the exfoliated mesothelial cells were not so obvious, and the expression of AQP-1 in peritoneal mesothelial cells was increased in AI group ( P 〈 0. 05 ). Conclusion AI can relieve the damage of mesothelial cells induced by high-concentration glucose peritoneal dialysate, enhance AQP-1 expression in peritoneal mesothelial cells, and improve the transcellular transport of peritoneal dialysate kept in the abdominal cavity at early stage, thereby enhancing the peritoneal water removal and increasing the amount of UF and Net UF.
