摘 要:【目的】观察诱生型一氧化氮合酶(iNOS)在脉冲噪声暴露白色和杂色豚鼠耳蜗中的表达,以探讨黑色素对耳蜗损伤的保护作用机理。【方法】将白色和杂色豚鼠暴露于脉冲噪声,复制耳蜗损伤的模型。采用免疫组织化学的方法观察iNOS在两组豚鼠耳蜗中的表达。【结果】iNOS在两组脉冲噪声暴露豚鼠耳蜗中均呈阳性表达,以血管纹和螺旋神经节的表达较强,而且iNOS在白色豚鼠耳蜗的表达显著强于杂色豚鼠。【结论】耳蜗黑色素拮抗耳蜗损伤的作用可能是通过抑制耳蜗iNOS表达来实现的。Objective The expression of inducible nitric oxide synthase (iNOS) was investigated in the cochlea of white and pigmented guinea pigs exposed to impulsive noise to explore the contribution of melanin in protecting cochlea from damage. Methods Both white and pigmented guinea pigs were exposed to the impulsive noise to establish the animal model of cochlea injury. The expression of iNOS in the cochlea was examined by the method of immunohistochemistry. Results There showed a positive expression of iNOS in the cochlea of white and pigmented guinea pigs exposed to impulsive noise, and the iNOS expression level in the area of stria vascularis and spiral ganglion was higher. The white guinea pigs had a higher iNOS expression level in the cochlea than the pigmented guinea pigs. Conclusion The mechanism of melanin in counteracting cochlea injury induced by impulsive noise is probably related with the inhibition of iNOS expression in the eochlea.
