清热祛湿方对2型糖尿病胰岛素抵抗模型大鼠TNF-α影响的实验研究

摘　　要：目的研究清热祛湿方对2型糖尿病模型大鼠肿瘤坏死因子-α(TNF-α)的影响,以观察清热祛湿方治疗对胰岛素抵抗(IR)的影响。方法雄性Wistar大鼠90只,采用高脂饲料喂养联合腹腔注射链脲佐菌素(STZ)的方法,建立2型糖尿病IR大鼠模型,取50只造模成功的大鼠,与正常组一起纳入本研究。模型大鼠随机分为5组,分别为清热祛湿方低剂量组、中剂量组、高剂量组,二甲双胍组,模型组。于给药2月后,各组大鼠治疗前后分别测定TNF-α、空腹血糖(FBG)、空腹胰岛素(FINS)、血脂,计算胰岛素敏感指数(IAI)、胰岛素抵抗指数(HOMA-IR)。结果清热祛湿方可明显降低模型大鼠血清TNF-α水平,同时有降低血糖、血脂作用,治疗后IAI、HOMA-IR均明显改善,与对照组相比有统计学意义。结论清热祛湿方治疗可明显降低2型糖尿病IR模型大鼠TNF-α水平,增加胰岛素敏感性,改善IR。而且随着剂量的增大,疗效更加显著。Objective To investigate the effects of Qingrequshi decoction on tumor necrosis factor α（TNF - α）in insulin r .sistan rats with type 2 diabetes mellitus（T2DM）. Methods Ninety male Wistar rats were selected. Model rats with insulin resistance were established by feeding with high fat forage combined with intraperitoneal injection of small dose of streptozotocin. Fifty rats were rau domly assigned to 5 groups：Qingrequshi decoction high dosage group, medium dosage group and low dosage group treated with Qingrequshi decoction,control group treated with metformin,model group and normal group treated with distilled water for 2 months. The TNF- α, blood lipid,fasting plasma glucose and fasting insulin were observed before and after treatment. Insulin resistance was evaluated by IAI and HOMA - IR. Results After treatment,the levels of TNF - α,blood lipid,fasting plasma glucose and fasting insulin were reduced obviously while IAI and HOMA - IR were improved obviously in Qingrequshi decoction group. The difference was significant corn pared to control group. Conclusion Qingrequshi decoction could reduce the blood TNF - α,improve the metabolic disorder and ameliorate the degree of insulin resistance in insulin resistant model rats with T2DM.
