摘 要:为了探讨温度和液相保存时间对猪精子顶体酶活性的影响,采用紫外分光光度计检测猪精子顶体酶活性。结果显示:(1)鲜精、冷休克和热休克每106精子顶体酶活性分别为5.38、5.20和5.16 mIU,差异不显著(P>0.05)。(2)鲜精在4、17、25、30、37和39℃时每106精子顶体酶活性分别为4.29、5.01、5.03、5.17、4.78和4.61 mIU,随着温度的升高精子顶体酶活性逐渐提高,当温度达到30℃时达到最高,且4℃和30℃时差异显著(P<0.05)。(3)液相精液在4℃保存超过1 d时精子顶体蛋白酶活性显著下降(P<0.05);在17、25和39℃保存超过2 d时顶体酶活性显著下降(P<0.05),且在17℃保存效果较好。结论是猪精子顶体蛋白酶活性受温度的影响,在17℃的条件下3 d内保存的猪精子顶体酶活性最佳。The purpose of this study was to explore the temperature and storage time on the pig sperm acrosin activity,which was detected by UV. The results showed that : ( 1 ) Fresh, cold shock and heat shock sperm acrosin activities were 5.38, 5.20 and 5. 16 mIU per 106 sperm, respectively, but the difference was not significant. (2) Fresh sperm acrosin activities at 4℃, 17℃, 25℃, 30℃, 37℃ and 39℃ were 4. 29, 5.01, 5.03, 5.17, 4. 78 and 4.61 mIU per 106 sperm, respectively. Sperm acrosin activity increase along with temperature increasing. When the temperature reached 30℃, acrosin activity reached the highest, the difference was significant between 4℃ and 30℃ (P 〈 0.05). (3) When the preservation of more than 1 d at 4℃, the sperm acrosin activity decreased significantly (P 〈 0.05 ) ; when the preservation of more than 2 d at 17℃ ,25℃ and 39℃ ,acrosin activity decreased significantly (P 〈 0. 05 ). The efficiency of preservation at 17℃ was best. In conclusion, the temperature affects the pig sperm acrosin activity,and the temperature of liquid semen Preservation was suitable for 17℃. Under this temperature the pig sperm can be saved for 3 d.
