摘 要:目的观察GSK3特异性抑制剂氯化锂(LiCl)对去卵巢(OVX)大鼠骨密度的影响并探讨其作用机制。方法24只雌性SD大鼠,随机均分为3组:假手术(Sham)组、OVX组和LiCl组。Sham组仅切除一段肠系膜,OVX组和LiCl组行双侧卵巢摘除,LiCl组给予LiCl溶液140mg/(kg.d)灌胃,Sham组与OVX组均用生理盐水(1ml/d)灌胃,持续10周。每周称量大鼠体重,10周后观测各组大鼠骨密度、子宫重量及子宫内膜的改变;采用免疫组化和Westernblotting观察骨组织形态学改变和β-catenin蛋白的表达。结果10周后,与OVX组比较,LiCl组大鼠体重增幅小而骨密度有所增加,子宫内膜未出现萎缩性改变,骨小梁形态结构基本完整,骨小梁形态结构破坏少。与Sham组比较,OVX组成骨细胞β-catenin蛋白表达降低,而LiCl组表达明显升高。结论LiCl可使OVX大鼠的骨密度增加,可抑制由雌激素缺乏引起的子宫萎缩,从而有效预防雌激素缺乏导致的骨质疏松。Objective To investigate the effect of lithium chloride(LiCl)on bone mineral density in ovariectomized(OVX)rat,and study the possible mechanisms.Methods Twenty-four 3-month-old female SD rats were randomized into 3 groups(8 each):sham group,ovariectomized(OVX)group and LiCl group.Animals in OVX and LiCl group were bilaterally ovariectomized,and those in sham group were treated with partial mesentery resection.Rats in LiCl group received a 140mg/kg dose of LiCl by gavage each day,and those in the two other groups were given 1ml of saline by gavage.The body weight of rats in all the 3 groups was weighed every week.Ten weeks later,all rats were sacrificed,and the bone mineral density(BMD),uterine weight and the changes in endometria were detected.Immunohistochemical staining and Western blotting were used to observe the histological changes in bone tissue and to detect the expression of β-catenin protein.Results The results of present experiment showed that,after ten weeks,the body weight increased obviously in OVX group(P〈0.01),while no significant increase was observed in LiCl group(P〉0.05)compared to that in sham group;the bone strength was enhanced significantly in LiCl group compared to that in OVX group(P〈0.01),and the treatment of LiCl prevented trabecular microarchitecture from deterioration and the endometrium from atrophy.The mechanism might be associated with the prevention of BMD from further decreasing,or even accompanied with some improvement in microarchitecture.The expression of β-catenin protein was decreased in OVX group compared with that in sham group,while increased significantly in LiCl group compared with that in OVX group.Conclusion LiCl could increase BMD of ovariectomized rat,prevent uterus from atrophy induced by lack of estrogen,thus efficiently preventing osteoporosis.
