摘  要:目的:通过测定大鼠动情周期、卵巢和子宫指数、性激素水平及透射电镜下观察卵巢超微结构的改变,探索生殖功能低下大鼠模型的制备方法,进而为临床治疗此类疾病提供可靠的实验依据。方法:采用12周龄SD雌性大鼠20只,随机分为正常组和模型组,模型组借鉴前期研究中小鼠的造模剂量和时间,采用雷公藤多甙40mg/(kg·d)灌胃,连续10周,正常组不给药。分别观察2组大鼠动情周期、性激素、子宫和卵巢指数,并观察卵巢超微结构的改变。结果:与正常组相比,模型组大鼠动情周期紊乱,表现为动情周期缺失、不完整或延长,血清E2/P水平明显下降,子宫及卵巢质量下降且卵巢组织结构受损。结论:胃饲雷公藤多甙40mg/(kg·d)10周能损伤大鼠生殖功能,造成雌性大鼠生殖功能低下。Object: Explore the preparation methods of reproductive dysfunction-rat-model, furthermore to provide reliable experimental basis for the clinical treatment of these diseases by measuring the rat estrous cycle, ovarian and uterine index, sex hormone levels and observing the ultrastructural changes of ovary under transmission electron microscope.Methods: 20 12-week-old SD female rats were randomly divided into normal group and model group.In the model group, we draw on preliminary studies in mice dose and time of the TWP 40 mg/ (kg·d)gavage for 10 consecutive weeks, while the normal group were not administered.The estrous cycle, sex hormone, uterus and ovaries index and the ultrastructural changes of the ovary were observed of the two group rats respectively.Results: In the model group, the estrous cycle was disorder manifested in lack of estrous cycle, incomplete or extended, serum E2/P levels were significantly decreased,the quality of the uterine and ovarian declined and the ovarian tissue structural damaged compared with the normal groups.Conclusion: Gastric feeding TWP 40 mg/ (kg·d) for 10 weeks can injure the reproductive function of the rats, resulting in reproductive hypofunction in female rats.
