摘 要:用免疫手术的方法分离白色(昆明白)或棕色[昆明白×C57BL/6N)F_1,自交20代以上]小鼠胚泡的内细胞团,通过显微注射术将分离出的内细胞团,注入另一品系(棕色或白色)小鼠胚泡的胚泡腔内,经体外培养后移植到假孕受体子宫内,生下了嵌合体小鼠。共注射受体胚泡137枚,注射后成活121枚,成活率为88.3%(121/137),将98枚成活的胚泡移植到14只同步假孕受体小鼠子宫,移植后成活受体11只,其中7只受体受胎,受胎率为63.6%(7/11),7只受胎的受体小鼠,共接受胚胎49枚,产仔鼠30只,产仔率为61.2%(30/49)。到目前为止共存活14只仔鼠,仔鼠存活率为46.6%(14/30)。在9只已生长出毛的仔鼠中,2只仔鼠属于毛色嵌合,新生仔鼠中另2只仔鼠的眼睛颜色与供体胚的品系相同。眼球与其它组织的嵌合性,待进一步分析,免疫手术可以快速准确地得到大量的纯内细胞团,这种方法与胚泡注射法相结合,为制备动物嵌合体提供一条有效的新途径。Mouse chimeras were produced by injecting the inner cell mass(ICM) into the blastocoeles of the recipient blastocysts and transferring the injected blastocysts into the uteri of recipient mice. The ICMs isolated by immunosurgery were obtained from White(Kongmlng White) or brown [(Kongming White×C57BL/6N) F_1, inbreeding over 20 generations] mice and injected into the blastocysts obtained from other strain of mice (brown or white). Among 137 injected blastocysts 121 blastocysts survived. The survival rate was 88.3% (121/137), 98 survived blastocysts were transferred into uteri of 14 synchronic pseudopregnant female mice. After transfer, 11 recipients survived. 7 recipients became pregnant. The pregnant rate was 63.6% (7/11). The 7 pregnant recipients received 49 embryos and gave birth to 30 offsprings. Survival rate of transferred embryos was 61.2% (30/49). Up to now 14 offsprings survived. The survival rate was 46.6% (14/30). Two of 9 offsprings in which coat colour can be distinguished already, were chimeras, and other 2 newly-born mice present the eye colour which like their parents of donor embryos. The chimerism of eye and other tissues will be checked in the near future. The method of immunosurgery can produce large number of pure ICMs quickly and curiously. This method combined with that of blastocyst injection provided a new efficient way for producing animal chimeras.
