驱铅丸对慢性铅接触大鼠肾脏氧化损伤保护作用的研究

摘　　要：[目的]从脂质过氧化损伤阐述中药驱铅丸对慢性铅中毒大鼠肾保护作用的可能机制。[方法]在饮水中添加0.02%醋酸铅,连续60d以制作大鼠慢性铅中毒模型,然后将造模动物随机分为4组:中药高剂量治疗组(中药驱铅丸按3.5g.kg-1.d-1的剂量灌胃)、中药低剂量治疗组(驱铅丸按0.7g.kg-1.d-1的剂量灌胃)、EDTA治疗组(依地酸钠钙加普鲁卡因肌肉内注射,50mg.kg-1.d-1,连续注射4d为1个疗程,休息4d后进行下一个疗程,连续治疗7个疗程)及模型组(不予任何治疗),连续60d后观察治疗前后血、尿及肾脏的部分元素含量变化,并检测谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、丙二醛(MDA)。[结果]经统计,与模型组比较,各治疗组尿铅、血铅含量显著降低(P﹤0.05),尿钙、尿锌降低,肾钙、肾锌增高(P﹤0.05),肾组织MDA降低,GSH增高(P﹤0.05,P﹤0.01),SOD变化没有显著性差异(P﹥0.05)。[结论]用于治疗慢性铅中毒的富锌中药驱铅丸能降低血液及肾组织铅含量,促进铅的排出;驱铅丸通过抗氧化损伤而发挥对铅造成的肾组织损害的保护作用。[Objective] To study the protection mechanism on lipid overoxidation injure of kidney in rat with chronic lead poisoning by Chinese traditional medicine QQP（Quqian Pill）.[Methods] Built up the chronic lead poisoning model by drinking 0.02% lead acetate for 60 days,then the rats were randomly divided into 4 groups：the low QQP dosage group（the rats were fed by QQP with 2.0 g.kg-1.d-1）,the high group（3.5 g.kg-1.d-1）,the EDTA group（by edetate calcium disodium with 50 mg.kg-1.d-1）,and model group without any treatments.Otherwise,the control group was 8 rats drinking distilled water.After 60 days,checked the changes of some elements（Pb,Zn,Ca） in the blood,urine and kidney,and the concentration of GSH.[Results] Compared with the model group,the lead of other groups in blood,kidney and urine was decreased significantly（P﹤0.05）,the concentration of weight and hemoglobin was increased（P﹤0.05）,calcium and zinc in urine fall down（P﹤0.05）,and calcium and zinc in kidney and blood was increased（P﹤0.05）,MDA was decreased（P﹤0.05）,GSH was increased（P﹤0.01）.[Conclusion] QQP can reduce the lead level in the blood and kidney,promote elimination of lead,it protects the kidney through anti-oxidation injure.
