摘  要:目的检测神经上皮干细胞与壳聚糖纤维的生物相容性,并探讨其在壳聚糖纤维材料上的生长、分化情况。方法从胚胎神经管取材神经上皮干细胞,并与壳聚糖纤维进行体外共培养。采用MTT法检测细胞在壳聚糖纤维上的活性,采用光镜和扫描电镜技术观察细胞在壳聚糖纤维上的存活、生长状况,采用免疫细胞化学染色鉴定神经上皮干细胞的分化情况。结果神经上皮干细胞可以贴附在壳聚糖纤维表面生长,并可分化为神经元和神经胶质细胞,细胞活性与单纯神经上皮干细胞培养组没有明显差别。结论壳聚糖纤维与神经上皮干细胞有良好的生物相容性,二者在外周神经损伤的治疗中具有良好的应用前景。Objective To explore the biocompatibility between neuroepithelial stem cells (NEPs) and chitosan fibers,and to observe whether the NEP stem cells are able to proliferate and differentiate on chitosan fibers. Methods Neural-tube-derived NEPs were seeded on chitosan fibers in vitro. MTT assay was used to determine the biocompatibility of chitosan fibers. A light and scanning electronic microscope was performed to examine the growth and survival. Immunocytochemical staining was used to examine the differentiation of NEPs. Results NEPs survived well on the surface of chitosan fibers,furthermore,they firmly attached to the chitosan fibers and differentiated into neurons and glia. Conclusion Chitosan fibers have excellent biocompatibility with NEPs,which offers a possible substitute for repairing peripheral nerve injury.
