摘 要:目的建立高效液相色谱法,改进注射用水溶性维生素中生物素的含量测定方法。方法色谱柱为COSMOSILC18(250×4.6mm,5μm),以缓冲液(取0.5g高氯酸钠,加入1ml磷酸,用水稀释至1000ml)为流动相A,乙腈-甲醇(1:2)为流动相B,以流速1.5ml/min进行梯度洗脱;柱温为30℃;检测波长为200nm。结果生物素在1.5~4.5μg/mg的浓度范围内呈线性关系,r=0.9995(n=5)。平均回收率为101.0%,RSD为0.6%(n=9)。结论本方法简便、准确,不受其他成分的干扰,可用于测定注射用水溶性维生素中生物素的含量。Objective To establish a HPLC method, to improve the content determination of biotin in water-soluble vitamins for injection. Methods COSMOSIL C18 (250mm × 4.6mm,5μm)was used as the column. Mobile phase A:buffer solution, Mobile phase B :aeetonitrile-methanol(1:2). The flow rate was 1.5ml/min, following the gradient elution. The column temperature was 30℃ and the detection wavelength was 200nm. Results Biotin presented a good linear in the range of 1.5 ~ 4.5μg/mg,r =0.9995(n =5). The average recovery was 101.0% ,RSD =0.6% (n =9). Conclusion The present method is simple and accurate for content determination of biotin in water-soluble vitamins for injection, and it also has the advantage of preventing the interference from other compounds.
