摘 要:目的利用BN大鼠建立β乳球蛋白致敏的动物模型。方法采用连续灌胃及腹腔注射致敏蛋白的方式致敏BN大鼠,ELISA测定β乳球蛋白特异性IgE水平。口服给予β乳球蛋白激发过敏反应,苏木素-伊红及甲苯胺蓝染色观察肠粘膜组织病理变化,计数肠浸润肥大细胞数及肥大细胞脱颗粒比例。结果模型组血清特异性IgE水平升高,β乳球蛋白激发后引起肠道炎症反应,肠粘膜肥大细胞浸润增加及肥大细胞脱颗粒比例增加。结论本实验建立的的BN大鼠模型是较理想的食物过敏模型。Objective To develop an animal model that mimics human cow milk allergy.Methods Test protein was administered i.p and orally to male BN rats for consecutive 4 weeks every 7 days.In different intervals,blood samples obtained from the tail of rat were used for determination β-lactoglobulin specific IgE level by ELISA.After oral administration of β-lactoglobulin,small intestine was used for histopathologic examination.Changes were observed with either HE stain or toludine blue stain.The intestinal mast cells and the percentage of the granule releasing mast cells were counted.Resuts Compared with the controls,the β-lactoglobulin specific IgE level were higher,and oral ingestion of the protein challenged the allergic inflammation in small intestinal.The intestinal mast cells and the percentage of the granule releasing mast cells were increased in model group.Conclusion BN rat may be a suitable animal model for investigation of food allergy.
