摘  要:目的研究异黄酮苷元滴丸和片剂的药动学及相对生物利用度。方法以SD大鼠为实验动物,采用高效液相色谱法测定异黄酮苷元滴丸和片剂单剂量灌胃给药后大鼠的血药浓度变化。用3P97软件进行药代动力学数据处理,计算分析药代动力学参数和相对生物利用度。结果异黄酮苷元滴丸:AUC为46.30μg.min.mL-1,cmax为0.89μg.mL-1,tmax为18.03 min;片剂:AUC为36.01μg.min.mL-1,cmax为0.41μg.mL-1,tmax为31.43 min,滴丸相对于片剂的相对生物利用度为128%。结论异黄酮苷元滴丸、片剂均符合一室模型,滴丸较片剂具有较高的生物利用度。本研究为异黄酮苷元滴丸研制提供了一定的依据。Objective To investigate the pharmacokinetic and relative bioavailability of isoflavone aglycone dripping pills and tablets in healthy SD Rats,and evaluate the pharmacokinetic effects of the two dosage forms of isoflavone aglycone.Methods 36 SD rats were intragastrically administered with the suspension solution of isoflavone aglycone dripping pills and tablets at single dose,and changes in plasma concentration were determined by HPLC.Pharmacokinetic data were processed with 3P97 software and the pharmacokinetic parameters and relative bioavailability were calculated.Results For dripping pills of isoflavone aglycone:AUC=46.30 μg·min·mL^-1,Cmax=0.89 μg·mL^-1,Tmax=18.03 min;and for tablets of isoflavone aglycone:AUC=36.01 μg·min·mL^-1,Cmax=0.41 μg·mL^-1,Tmax=31.43 min.The relative bioavailability of dripping pills and tablets was 128%.Conclusion The pharmacokinetics of dripping pills and tablets of aglycone isoflavone were in accordance with single compartment model,and the relative bioavailability of dripping pills of aglycone isoflavone was higher.
