摘  要:目的研究冠心病血瘀证、非血瘀证与血小板功能蛋白相关性。方法纳入冠心病血瘀证组22例、冠心病非血瘀证组22例,采集血浆分离血小板,提取血小板蛋白,用荧光差异显示二维凝胶电泳筛选差异功能蛋白,基质辅助激光解析/电离-飞行时间质谱对血小板差异功能蛋白进行鉴定。对鉴定成功差异蛋白,Western-Blotting进一步验证。结果筛选,并质谱成功鉴定10个差异蛋白点。冠心病血瘀证组比对照组,血小板功能蛋白有2个:cDNA FLJ53327、cDNA FLJ43573fis表达增多;有8个:Isoform 2 of Integrin alpha-IIb、FGG50kDa protein、A26C1B ANKRD26-like、Actin-cyto-plasmic1、Actin-cytoplasmic 2、cDNA FLJ52842、Fibrinogen beta chain、Keratin type I表达减少。Fibrino-genβ成功验证。结论冠心病不同证侯与血小板功能蛋白表达水平明显相关,部分血小板功能蛋白可能是冠心病不同证侯的物质基础之一。Objective To study the correlation between Blood stasis /non-Blood stasis and platelet s function proteins in coronary heart disease. Methods Two groups were established which were Blood stasis of coronary artery disease group(included 22cases) and non-Blood stasis of coronary artey disease(included 22 cases). Vein blood were drawed. Platelet was separated from the blood samples. Platelet proteins were extracted from platelet solution. To screen the platelet differential function proteins with two-dimensional Fluorescence Difference Gel Electrophoresis, the screened differential proteins spots were identi- fied with Matrix-assisted laser desorption/ionization- time of flight-time of flight mass spectrometry. The identified proteins were authenticated further by Western-Blotting. Results 10 proteins were screened and identified with MALDI-TOF-TOF. cDNA FLJ53327 and cDNA FLJ43573 fis were expressed in Blood stasis of coronary artery disease group much more than that in non-Blood stasis of coronary artey disease group. Iso- form 2 of Integrin alpha- IIb,FGG 50 kDa protein, A26C1B ANKRD26-1ike,Actin-cytoplasmic 1 ,Actin-cy- toplasmic 2 ,cDNA FLJ52842, Fibrinogen beta chain and Keratin type I were expressed in Blood stasis of coronary artery disease group much less than that in non-Blood stasis of coronary artey disease group. Fibrinogen 13 was authenticated further by Western-Blotting. Conclusion Our experiment manifested the obivious correlation between the different Symptom complexs and the expression level of platelet function proteins. Partial platelet function proteins might be one of the reasons which induced the different Symptom complexs of coronary artery disease.
