摘 要:目的:比较瘦素(1eptin)不同给药方法在小鼠局灶性脑缺血/再灌注损伤中的保护作用。方法:雄性昆明小鼠60只,随机分成5组:假手术组、PBS对照组、腹腔注射Leptin 1mg/kg组、鼻腔滴入Leptin 0.5 mg/kg组和鼻腔滴入Leptin 1 mg/kg组。Leptin均于脑缺血时立即给予。线栓法制备小鼠局灶性脑皮质缺血/再灌注损伤模型,缺血2 h再灌注24 h后评价神经功能学状态,噻唑兰(TTC)染色观察脑梗死体积,分光光度法检测损伤侧脑组织匀浆中乳酸脱氢酶(LDH)和一氧化氮(NO)水平;苏木素-伊红(HE)染色观察脑组织病理学改变。结果:与PBS对照组比较,1eptin给药各组神经功能损伤均不同程度减轻,脑皮质梗死体积明显缩小,LDH水平明显降低,NO水平呈现升高趋势,其中以鼻腔给予1eptin 1mg/kg组对降低LDH的效果最为明显。结论:在对小鼠局灶性脑缺血/再灌注损伤的保护作用中,鼻腔给药方法简便、效果好,可作为首选给药方式。Objective:To compare the protective effect of administration of leptin in different ways in mice with focal cerebral ischemia/reperfusion injury. Methods: Sixty male Kunming mice were randomly divided into 5 groups: sham operation group,phosphate buffer solution (PBS) group, and three groups with rats given leptin in different ways: 1 mg/kg leptin intraperitoneal injection,0. 5 mg/kg or 1 mg/kg leptin given into nasal cavity. Focal cerebral ischemia was reproduced by middle cerebral artery occlusion (MCAO) for 2 hours and followed by reperfusion. After 24 hours of reperfusion,neurologieal deficit scores and the size of cerebral infarction were determined,the levels of lactate dehydrogenase (LDH) and nitric oxide (NO) in ischemia brain homogenate were monitored by spectrophotometry, HE staining was used to observe the pathological changes in hippocampus region of the ischemia reperfusion mice. Results:Compared with the PBS group, neurological deficits scores and the infaret volume of the three groups of mice receiving leptin were lessened in different extent, the level of LDH was decreased significantly, and NO level gradually elevated. The effects of administration 1 mg/kg leptin via nasal cavity were the best on decreasing the LDH level. Conclusion:The administration of leptin via nasal cavity may be a novel effective and convenient pathway to protect the brain tisse with ischemia/reperfusion injury.
