摘 要:目的探讨乳鼠窦房结细胞的分离纯化方法,并观察其形态学特征。方法取新生SD大鼠的窦房结组织,胰酶消化后采用速差贴壁法及5-溴脱氧尿嘧啶核苷(5-BrdU)纯化处理进行原代培养。对培养细胞进行形态学观察,用电生理技术记录细胞的动作电位。结果在培养的窦房结细胞中观察到三种不同形态的细胞,即梭形细胞、三角形细胞与不规则形细胞。其中梭形细胞所占比例最高,且形态结构与电生理特点符合窦房结起搏细胞的特点。三角形细胞与心房肌细胞特征相似。结论采用酶消化法结合速差贴壁及5-BrdU处理,可以显著提高窦房结梭形细胞的比例,是可靠的窦房结细胞的分离纯化方法。Objective To find a way of purification on sinoatrial node cells (SNCs) isolated from newborn rats and to observe the morphological characteristics. Methods The SNCs were isolated from sinus node tissues of rats by trypsin and purified cultured with differential attachment and 5 - bromodeoxyuridine (BrdU) treatment. The cultured cells were identified by morphology and were further confirmed by action potential measured with electrophysiological technique. Results Three different types of cells were observed among the purified cultured SNCs:spindle,triangular and irregular cells. The spindle cells took up the greatest proportion, and the configuration, structure and electrophysiological character of the spindle cells were accorded with the character of pacemaker cell of SNCs. Triangle cells isolated from SNA were similar with triangle cells of atrial muscle. Conclusions Isolation by trypsin combined with differential attachment and BrdU treatment is a reliable approach to culture sinoatrial node cells.
