摘  要:目的观察内毒素(LPS)对巨噬细胞的凋亡作用以及重组人生长激素(rhGH)的干预效果。方法巨噬细胞系U937细胞经传代培养24h后,换用含LPS(40μg/mL)和LPS+rhGH(4u/L)的新鲜培养液继续培养16h,流式细胞仪检测细胞凋亡,免疫细胞化学法检测巨噬细胞内NF-κB的活化程度。结果U937细胞经LPS和LPS+rhGH作用16h后,rhGH明显降低LPS诱导U937细胞的细胞凋亡率,并可抑制NF-κB的活化。结论rhGH能抑制LPS诱导的U937细胞凋亡,并对NF-κB的活化有一定的抑制作用。【Objective】The effect of recombinant human growth hormone (rhGH) on lipopolysaccharides (LPS) induced macrophage apoptosis and Nuclear factor-kappaB (NF-κB) activity were tested in vitro.【Methods】U937 macrophage cell line cells were treated with LPS (40μg/mL) or LPS with additional rhGH (4u/L) for 16 hrs in vitro.The apoptosis percentages of U937 cells were assessed flowcytometrically and the macrophage nucleus NF-κB expression evaluated immunohistochemically.【Results】The apoptotic percentages of U937 cells treated with LPS and additional rhGH was significantly reduced as compared with the cells treated with LPS alone (4.13±0.57% vs 9.1± 2.40%;P 〈0.01);while the level of NF-κB activation of LPS alone treated cells was significantly higher than that cells treated with additional rhGH (41.1±10.46% vs 9.3±1.9%;P 〈0.01).【Conclusions】These results suggest that rhGH can reduce the LPS induced apoptosis of U937 cells,which might be contribute to its inhibiting on NF-κB activation.
