摘 要:目的:研究组织工程皮肤与正常皮肤表皮层组织学的差异,并探讨其意义.方法:用组织工程方法构建人全层皮肤,以正常在体皮肤作为对照,组织学、免疫细胞化学、透射电镜观察表皮层的异同.结果:组织工程皮肤与正常皮肤在组织学上有一定差异,缺少上皮钉突,CK19在组织工程皮肤成熟期表达阴性,但CK10和pan-CK在两者表达皆为阳性,透射电镜观察上皮结构接近正常皮肤.结论:组织工程皮肤表皮层与真皮层结合不如正常皮肤紧密,可能与缺少上皮钉突有关,其表皮细胞基本分化成熟.AIM:To study the differences in histology of the tissue-engineered skin and normal human skin.METHODS:The bilayer skin replacement was constructed by tissue engineering method,compared with the normal human skin by histological and immunohistochemistry methods.RESULTS:There were some differences in tissue-engineered skin and the normal skin.Epidermis rete pegs were presented only in the normal skin,CK19 was negative in the matured tissue engineered-skin,but both CK10 and pan-CK were positively expressed in tissue-engineered skin and the normal skin.Epithelium structure by TEM was similar to the normal skin.CONCLUSION:The dermo-epidermal junction in tissue-engineered skin was less tighter than normal skin,may be due to the absence of epidermis rete pegs,epidermal cells had differentiated.
