摘 要:目的:探讨通过RNA干扰技术下调SOCS3表达防治高脂餐大鼠代谢紊乱的作用.方法:取SD6wk龄大鼠45只,随机分为3组:普通饮食对照组(n=15),下丘脑注射空白质粒,普通饲料喂养;高脂饮食对照组(n=15),下丘脑注射空白质粒,高脂饲料喂养;实验组(n=15),下丘脑注射shR-NA-SOCS3序列慢病毒重组体,高脂饲料喂养.每周测体质量1次.饲养8wk末,测空腹血糖和体质量,处死后立即取心脏血进行血清胰岛素、三酰甘油、总胆固醇、血清瘦素测定.结果:实验组体质量、三酰甘油、总胆固醇低于高脂饮食对照组(P<0.01),高于普通饮食对照组(P<0.05).实验组血清瘦素、血清胰岛素浓度低于高脂饮食对照组组(P<0.01),与普通饮食对照组比较无统计学差异.结论:下调SOCS3基因表达可以降低体质量、三酰甘油、血清胰岛素、总胆固醇及血清瘦素水平,对肥胖症、糖尿病及慢性并发症可能有预防和治疗作用.AIM:To study the effects of NRAi-mediated gene silencing of SOCS3 on metabolic disturbance in high-fat diet rats.METHODS:Forty five 6-week-old healthy Sprague-Dawley(SD) rats were adopted to our research and divided into 3 groups randomly.In the standard control group(n=15),the control lenti were stereotaxically injected into the hypothalamus of rats which then fed with standard chow;In the high-fat diet control group(n=15),the control lenti were stereotaxically injected into the hypothalamus of rats which then fed with high-fat diet;In experiment group(n=15),the SOCS3 lenti were stereotaxically injected into the hypothalamus of rats which then fed with high-fat diet.Animal weight was measured weekly.Up to 8 weeks after treatment,before exeduted,fasting plasma glucose was measured,After exeduted,serum was collected for measurement of serum leptin,insulin,triglyceride,total cholesterol.RESULTS:The experiment group rats gained smaller weight,lower triglyceride and total cholesterol than high-fat control group rats(P0.01)and showed larger weight,higher triglyceride and total cholesterol than CS rats(P0.05).Levels of serum leptin and insulin were lower in experiment group rats than high-fat control group rats(P0.01).There were no significant differences in comparison of levels serum leptin and insulin in experiment group rats with the standard control group rats.CONCLUSION:Body weight,serum leptin,insulin,triglyceride and total cholesterol concentration were decreased through down-regulation of SOCS3 expression by RNAi-mediated gene silencing of SOCS3,suggesting that down-regulation of SOCS3 expression is a promising therapeutic approach in obesity,diabetes and chronic complications.
