摘 要:目的:使用超声指标观察大鼠颈总动脉机械损伤模型,评价干细胞动员对其修复过程的影响,并与病理学指标对照分析。方法:将70只大鼠随机分为Ⅰ、Ⅱ、Ⅲ3组,Ⅰ组为假手术组,Ⅱ组为颈总动脉内皮机械损伤自然修复组,Ⅲ组为颈总动脉内皮机械损伤后干细胞动员组,使用粒细胞集落刺激因子动员干细胞。Ⅱ、Ⅲ组又根据手术时间各分为A、B、C 3个亚组,在术后1天、7天、28天分别超声观察颈总动脉管腔、血流速度、脉动指数(PI)和阻力指数(RI),与病理切片对照。结果:Ⅱ组大鼠颈总动脉内皮剥脱后管腔狭窄,血流速度减慢,PI、RI增高,均于损伤第28天达峰值,Ⅲ组大鼠颈总动脉内皮剥脱后管腔狭窄,血流速度减慢,PI、RI增高,7天与Ⅱ组比较无显著差异,28天时上述指标显著优于Ⅱ组,但仍较Ⅰ组差。结论:超声指标能够无创动态观察大鼠颈总动脉损伤及修复过程,使用干细胞动员可以显著加快修复。Objective:To observe the de-endotlielialization model in rat carotid artery with Doppler ultrasound and pathologic analysis, and to evaluate the effect of stem cell mogilization to this model. Methods:70 rats were randomly divided into Ⅰ , Ⅱ, Ⅲ 3 groups, Ⅰ group was given sham operation, Ⅱ group recovered naturally after being injured, Ⅲ group was hypodermic injected granulocyte colonystimulating factor(G-CSF) to mobilize the stem cell. The rat model was established by denuding the carotid endothelium, and dynamie changes of irmer diameters, bemodynantics, pulsatile index(PI) and resistance index(RJ) were observed at 1 d, 7 d and 28 d after deendothelialization with ultrasound, and were compared with pathology. Resadts: The luminal diameters, the maximal and minimal blood velocities were decreased after de-endothelialization, whereas the pulsatile index(PI) and resistance index(RI) were increased in beth Ⅱ and Ⅲ group, yet, 28 days after injured PI and RI were lower in Ⅲ group than those in Ⅱ group. Conclusion: Doppler ultrasonic technique can be used to observe the injury and repairation of rat carotid artery after de-endothelialization, and stem cell mobilization is useful to the recovery of artery after injured.
