摘 要:目的:探讨四君子汤对脾虚模型大鼠头孢羟氨苄药动学的影响。方法:24只大鼠随机分为4组:生理盐水组、生理盐水+四君子汤组、利血平组、利血平+四君子汤组。给予相应药物干预两周后,灌服头孢羟氨苄(60 mg/kg),给药后不同时间点采集血样,用高效液相色谱法分析头孢羟氨苄的血药浓度,根据血药浓度-时间数据计算药动学参数。结果:利血平引起的大鼠症状类似于中医临床脾虚证。头孢羟氨苄在生理盐水组、生理盐水+四君子汤组、利血平组、利血平+四君子汤组大鼠体内的药动学参数分别为:AUC(0~∞)=59.72±8.03,62.30±9.27,75.08±11.83和63.07±9.21 mg·h^(-1)·L^(-1);Cmax=25.21±2.47,24.45±4.20,34.04±4.77和24.15±5.38 mg/L。与生理盐水组相比,利血平组大鼠体内头孢羟氨苄的AUC(0~∞)和Cmax增加(P<0.05);与利血平组相比,利血平+四君子汤组大鼠体内头孢羟氨苄的AUC(0~∞)和Cmax降低(P<0.05),而生理盐水组与生理盐水+四君子汤组参数之间没有显著性差异。结论:脾虚状态下头孢羟氨苄的生物利用度有所增加,四君子汤可使其吸收恢复接近正常。Objective: To investigate the influence of "Sijunzi Decoction" on the pharmaeokinetics of Cefadroxil in rats of spleen deficiency. Methods: Twenty-four rats were randomly divided into four groups: NS ( normal rats given 0.9 % saline solution), NS + SJZT (normal rats given 0.9% saline solution and "Sijunzi Decoction" ), R (rats pretreated with Reserpine) and R + SJZT ( rats pretreated with Reserpine and then cured with Sijunzi Decoction). After two weeks of correspondent intervention, the rats were garaged with cefadroxil 60 mg/kg. Then, the blood samples were collected at different intervals and the concentrations of cefadroxil in plasma were determined by high-pressure liquid chromatography. Pharmacokinetic parameters were calculated accord- ing to the data of plasma concentration-time. Results: Reserpine led to the pattern similar to spleen deficiency of TCM in rats. The pharmacokinetic parameters of Cefadroxil in NS, NS + SJZT, R and R + SJZT groups were as follows: AUC (0 -∞ ) = 59.72 ± 8.03, 62.30 ± 9.27, 75.08 ± 11.83 and 63.07 ±9.21 mg·h^-1·L^-1 ,respectively , and Cmax=25.21± 2.47, 24. 45 ± 4.20, 34.04 ± 4.77 and 24.15 ± 5.38 mg/L, respectively. Compared with NS group, the AUC(0 - ∞ )and Cmax of Cefadroxil in R group were increased significantly(P 〈0.05). Compared with the R group, the AUC(0-∞ )and Cmax of Cefadroxil in R + SJZT group were decreased significantly (P 〈 0.05 ). However, there was no significant difference in the parameters between NS group and NS + SJZT group. Conclusion : The oral bioavailability of cefadroxil was increased in the condition of spleen deficiency pattern, whereas the "Siiunzi Decoction" can recover its absorption to normal state.
