摘  要:目的:研究维生素D3对人乳头瘤病毒(HPV)16型DNA-永生化的人宫颈上皮细胞(HCE16/3细胞)病毒基因表达的调节。方法:使用Northern印迹法分析了维生素D3或(和)维甲酸处理的HCE16/3细胞病毒早期基因表达。结果:在缺乏血清和生长因子的培养条件下,单独使用维生素D3或维甲酸并不明显改变病毒早期基因的表达,但维生素D3加维甲酸合并使用可以抑制细胞病毒早期基因的表达。结论:维生素D3加维甲酸合并使用可以抑制细胞病毒早期基因的表达。Objective:To study the effect of the active metabolism of vitamin D, i.e., 1,25-dihydroxycholecalciferol (1,25-D 3) on the expression of viral early genes of human papillomavirus (HPV) type 16 DNA-immortalized human cervical epithelial cells (HCE16/3 cells).Mothods:The effect of 1,25-D 3 and retinoic acid, alone and in combination, on the expression of viral early genes in cultured HCE16/3 cells was determined by Northern blot.Results:Vitamin D 3 or retinoic acid alone had little effect on the expression of viral early genes of HCE16/3 cells. However, the expression of viral early genes in HCE16/3 cells was inhibited with vitamin D 3 in combination with retinoic acid.Conclusion:Vitamin D 3 inhibits the expression of viral early genes in HPV DNA-immortalized human cervical epithelial cells in combination with retinoic acid. It may provide the experimental base for treating HPV-associated tumors with vitamin D 3 and retinoic acid in the future.
