摘  要:目的对左氧氟沙星在治疗大鼠肺炎感染模型中的不同给药方案进行比较。方法40只被大肠埃希菌标准菌株(ATCC25922)感染为肺炎的SD大鼠,分成两组,按照不同给药方案给药,组1为腹腔给药4.5 mg.kg-1,B id;组2为腹腔给药9mg.kg-1,Qd。微生物法测其血药浓度。根据各组中的左氧氟沙星的药动-药效学参数,评价不同给药方案的疗效。结果组1的平均血药峰浓度(2.56±0.02)明显低于组2(4.29±0.03),平均血药最低浓度(0.42±0.01)高于组2(0.29±0.01)。两组治疗后的肺组织匀浆细菌培养结果显示其细菌清除率分别为82%和83%,经统计学处理差异无显著性。结论两组给药方案治疗下呼吸道感染疗效均较好,效果差异性无显著性。但组2血药峰浓度高,给药次数少,更方便治疗。Aim To compare different dosages of levofloxacin in treating pneumonia of rat model. Methods 40 SD rats which were infected with standard strains of Escherichia eoli (ATCC25922) for pneumonia were divided into two groups, according to the administration of different dosages. The rats of Group 1 were injected drug by Intravenous 4.5 mg·kg ^- 1 , Bid. The Group 2 were injected drug by Intravenous 9 mg·kg^-1 , Qd. The microbiological assay methods were used for determination of levofloxaein in serum. According to the group of levofloxacin pharmacokinetic - pharmacodynamie parameters the effects of different drug dosage were evaluated. Results The study showed that the average peak concentration of group 1 ( 2.56 ±0.47 ) was significantly lower than that of group 2 (4.29 ± 0.95 ), but the average tough concentration of group 1 (0.42 ± 0. 18 ) was higher than that of group 2 (0.30 ± 0. 19 ). After treating pneumonia of the rats model, the bacterial culture results of lung tissue homogenate show the bacterial clearance rates were 82% and 83% respectively. There were no significant differences between the two groups after statistic analysis. Conclusions The two drug-administration programs are all ideal to treat the respiratory tract infection. There was no significant difference in efficacy in two groups. But the peak level of levofloxacin was higher, the number of infusion was less, more convenient treatment in group 2.
