摘  要:目的观察大鼠局灶性脑缺血后脑源性神经营养因子(BDNF)及神经生长因子(NGF)含量的变化,研究加减补阳还五汤对大鼠局灶性脑缺血后脑内BDNF及NGF含量变化的影响,探讨其对缺血性脑损伤的神经元保护机制。方法用大鼠大脑中动脉线栓法(MCAO)建立脑缺血模型,采用免疫组化方法观察BDNF及NGF在不同时间点的表达。结果与假手术组相比,模型组及治疗组大鼠BDNF及NGF的表达在各时间点均增强,以缺血48 h增强最为显著,且治疗组大鼠比模型组大鼠表达增强更显著。结论脑缺血后神经元内BDNF和NGF的含量增多,有利于受损神经元的恢复。加减补阳还五汤可能通过促进BDNF、NGF的表达而保护神经元。Objective To observe the changes in the expressions of brain - derived neurotrophic factor(BDNF) and nerve growth factor (NGF)after focal cerebral ischemia in rats and study the impacts of modified Bu Yang Huan Wu Tang on the changes in BDNF and NGF concentration after focal cerebral ischemia as well as explore its neuroprotective mechanism in ischemic brain injury. Methods The model of foeal cerebral ischemia was established by middle cerebral artery, occluding(MCAO). IHC method was adopted to observe the expressions of BDNF and NGF at different time spots. Results Compared with sham - operated group,the expressions of BDNF and NGF increased at different time spots in either model group or treatment group. The increase was the most significant in 48 h ischemia and the increased expression in treatment group was much more significant than that in model group. Conclusion The increase of BDNF and NGF in neurons after focal cerebral ischemia benefits the recovery of injured neurons. Modified Bu Yang Huan Wu Tang may protect neurons by promoting the expressions of BDNF and NGF.
