摘 要:目的:观察中药对糖尿病大鼠蛋白非酶糖化、肿瘤坏死因子α(TNF-α)及尿清蛋白的影响,探讨其对糖尿病肾病(DN)的防治作用及可能的作用机制。方法:雄性Wistar大鼠30只,随机平均分为3组,正常对照组、糖尿病对照组、糖尿病牛蒡[10ml/(kg·d)]治疗组,腹腔注射链脲佐菌素(60mg/kg)诱发糖尿病。于4、8、12、16周测尿清蛋白,16周后,取血测血糖、TNF-α,处死大鼠,分离肾脏,测定组织非酶糖化。结果:糖尿病组大鼠尿清蛋白有上升趋势,牛蒡治疗组尿清蛋白基本维持不变,牛蒡治疗组蛋白非酶糖化受抑制,TNF-α表达下降。结论:牛蒡可以降低尿蛋白,减轻肾脏损害,其防治DN的机制可能与抑制蛋白质非酶糖化,减少细胞因子的产生有关。Objective: To observe the effects on protein non-enzymatic glycation in diabetic rats, tumor necrosis factor-α (TNF-α) and urine albumin, and to explore the development of diabetic nephropathy (DN) of the preventive effect and possible mechanism of action. Methods: Male Wistar 30 rats were randomly divided into three groups, normal control group, diabetes control group, group diabetic rats treated with arctium Lappa L [(10 ml/kg·d); intraperitoneal injection of streptozotocin (60 mg/kg)induced diabetes. Urine protein measured at 4,8,12,16 weeks, 16 weeks later, blood glucose measurement, TNF-α, rats were killed, separation kidney, measured organizations, non-enzymatic glycation. Results: The diabetic rats urinary albumin upward trend, arctium Lappa L treatment of urinary albumin essentially unchanged, arctium Lappa L treatment group protein non-enzymatic glycosylation was inhibited, TNF-α expression decreased. Conclusion: arctium Lappa L can reduce urinary protein, to reduce kidney damage, its prevention and control mechanism of DN may inhibit non-enzymatic glycation of protein, reducing the production of cytokines.
