Effect of Electro-acupuncture on 5-HT_(1A) Receptor of Hippocampus of the Rat Model with Chronic Stress Depression

摘  要:目的:在建立慢性不可预见性应激抑郁大鼠模型的基础上,采用3H放射性配体受体结合分析法,测定大鼠海马5-HT1A受体特异性结合量,探讨电针抗抑郁机制。方法:将45只SD大鼠随机分为正常对照组、模型组和电针组,每组15只。用慢性不可预见性轻度刺激结合孤养的方法造模。造模同时给予电针治疗。穴位:选取百会穴、太冲穴,频率2Hz,电流强度为0.6mA左右,留针20min,每日1次,连续针刺21d。结果:正常对照组和模型组大鼠海马匀浆粗膜的[3H]8-OH-DPAT特异性结合分别为(30.16±3.34)fmol/mgprotein和(19.42±2.12)fmol/mgprotein。抑郁大鼠海马匀浆粗膜的[3H]8-OH-DPAT特异性结合明显低于正常对照组(P<0.05)。电针组大鼠经电针治疗21d后,海马匀浆粗膜的[3H]8-OH-DPAT特异性结合为(28.53±1.66)fmol/mgprotein,明显高于模型组(P<0.05),与正常对照组没有显著性差异。结论:抑郁大鼠海马5-HT1A受体的结合位点明显下降,电针治疗可以显著提高抑郁大鼠海马5-HT1A受体的结合位点。Objective: Based on the establishment of rat models of chronic unpredictable mild stress depression, 3H radioligand receptor binding assay was used to measure the specific binding capacity of 5 - HT1A receptor in rat hippecampus, to explore the anti-depressant mechanism of electroacupuncture. Methods: 45 SD rats were randomly divided into normal control group, model group and electro-acupuncture group( n = 15 ). The method of chronic unpredictable mild stimulation together with solitary feeding was used to make model rats. At the same time, electrical acupuncture treatment was given in modeling. Selecting acupeints : Baihui and Taichong were selected to give frequency 2 Hz, current intensity of 0.6 mA or so, leaving needle 20 min, one time a day, continuous needlling for 21 d. Results : [^3H ] 8 - OH - DPAT specific binding of hippocampal homogenate thick membrane of the normal control group and the model group were respectively ( 30. 16 ± 3. 34) fmol/mg protein and ( 19. 42 ± 2.12) fmol/mg protein. [ ^3H ] 8 - OH - DPAT specific binding of hippocampal homogenate thick membrane of depression rats was significantly lower than that of the control group (P 〈 0.05). [ ^3 H ] 8 - OH - DPAT specific binding of hippocampal homoge- hate thick membrane of the electro-acupuncture group after 21 days' electro-acupuncture treatment was (28.53 ± 1.66) fmol/mg protein, significantly higher than that of model group ( P 〈 0.05 ), there was no significant difference comparing with the normal control group. Conclusion: The binding sites of hippocampal 5 - HT1A receptor in the depression rats decreased significantly, electro-acupuncture treatment can significantly improve the binding sites of hippocampal 5 - HT1A receptor in the depression rats.
