摘  要:从河北唐山分离到一株疑似猪繁殖与呼吸综合征病毒,接种Marc-145细胞,经过4代盲传后,出现细胞病变,经鉴定为PRRSV,命名为TS株。根据GenBank公布的PRRSV JXA1株ORF3基因的核苷酸序列,设计并合成一对特异性引物,用RT-PCR方法扩增完整ORF3基因,将扩增产物连接到pMD19-T载体并转化克隆菌,将阳性重组质粒PMD-GP3进行序列测定与分析。应用DNA Star软件,将测序结果与国内外已发表野毒株和疫苗株的ORF3基因进行序列比较,并绘制系统进化树。这为研制PRRSV新型疫苗及诊断试剂奠定了基础。One PRRS virus strain TS was isolated from pigs ofHebei Tangshan after inoculated to Marc-145 cell. The full-length of ORF3 gene of PRRSV strain TS was amplified by RT-PCR with a pair of specific primers designed according to the ORF3 sequence of PRRSV JXA1 strain, and then cloned into plasmid pMD19-T. The recombinant plasmid named as pMD-GP3 was sequenced and analyzed. The sequence was compared with that of other PRRSV sWains in GenBank by using DNAStar software and the phylogenetic tree was drawn. It laid the foundation for further study for genetic engineering vaccine and diagnostic reagent for PRRSV.
