中药复方“松友饮”对高转移人肝癌裸鼠原位移植瘤的抑制作用

摘　　要：目的:研究中药"松友饮"对高转移人肝癌裸鼠原位移植肿瘤生长、转移和生存期的影响。方法:以肝癌细胞(MHCC97H)细胞建立裸鼠移植瘤模型,32只成模裸鼠随机分成对照组C0和"松友饮"剂量递增的C1、C2和C3组,肝脏接种肿瘤24h内开始1次/d灌胃给药,连续5周,每周测体质量。第35d处死裸鼠,计算肿瘤体积及转移情况。另选12只成瘤裸鼠分成对照组和"松友饮"组,选择上述结果中最合适的剂量,观察生存期。结果:C0、C1、C2和C3组肿瘤体积分别为(2072.73±801.23)mm3、(1779.48±969.59)mm3、(976.30±608.01)mm3和(959.99±376.47)mm3,C2、C3与C0相比差别有统计学意义;C2组抑瘤率52.90%,肺转移程度明显下降。按C2组剂量给药,裸鼠生存期(75.00±3.86)d,对照组(52.00±2.34)d(P<0.01)。结论:连续5周"松友饮"灌胃抑制HCC裸鼠模型肿瘤生长、转移,延长荷瘤裸鼠生存期,对HCC病人有潜在的应用价值。Objective： To investigate the effects of Songyouyin on tumor growth, metastasis and survival in nude mice bearing human hepatocellular carcinoma （HCC） xenografts. Methods： Metastatic human HCC nude mice models were established by orthotopic implantation with a high metastatic potential human HCC cell line MHCC97H. Thirty-two nude mice bearing orthotopic xenografts were randomized into control group CO and intervention groups C1, C2 and C3 with stepwise increased dosage of Songyouyin. Daily intragastric adminstration of Songyouyin was given for 5 consecutive weeks 24 hours after orthotopic implantation. Body weight was recorded weekly. Mice were sacrificed on day 35th, tumor volume and metastasis were examined. Another 12 mice beating human HCC xenograft were randomized into control group and Songyouyin group to observe life span. The dosage of Songyouyin was selected according to the result of the above mentioned experiment. Results： The tumor volume was（2072.73 ± 801.23）mm^3,（1779.48± 969.59）mm^3,（976.30 ± 608.01）mm^3 and（959.99 ± 376.47）mm^3 in C0, C1, C2 and C3 group respectively with significant statistical difference,when C2 and C3 compared with C0.Tumor inhibition rate in C2 group was 52.90%. The lung metastatic extent （grade） reduced significantly in group C2 when compared with control group Co （P〈0.05）. Life span of mice in Songyouyin group was significantly longer than that in the control, being（75.00±3.86）days versus（52.00 ±2.34）days. Conclusion： Five consecutive weeks administration of Chinese herbal compound prescription of Songyouyin resulted in inhibition of tumor growth and metastasis, and extended life span of nude mice bearing human HCC xenografts with high metastatic potential, indicated its potential use in patients with advanced HCC.
