摘  要:目的研究针刺治疗对脑梗死大鼠血清蛋白的影响。方法雄性SD大鼠20只,线栓法建立脑梗死模型,其中对照组10只,针刺组10只。针刺后1周,取血清,二维电泳分离蛋白,PDQ软件寻找差异表达的蛋白,用MALD I TOF/TOF串联质谱仪分析蛋白质,所得的质谱图通过GPS软件在MASCOT数据库中检索并鉴定蛋白质。结果针刺后血清中有13种蛋白的表达下调,2种蛋白表达上调。选取3种蛋白进行鉴定,针刺后表达下调的蛋白是血清淀粉样蛋白P,表达上调的是丝氨酸蛋白酶抑制剂和白蛋白。结论针刺可能通过提高丝氨酸蛋白酶抑制剂的表达和肝合成蛋白的功能对脑缺血起保护作用。Objective To study the effect of acupuncture on the expression of serum proteins in rats with cerebral infarction. Methods Twenty SD adult male rats were randomly assigned into control group (n = 10), and aeupunture group (n = 10 ). The cerebral ischemia model was estabished by using suture -occluded method. After being treated for 1 week by acupunture, rats were sacrificed. Two - dimensional ( 2 - D) gel electrophoresis was applied to separate serum proteins. PDQuest software was used to detect the differential expression of proteins between control and acupuncture groups based on 2 - D protein mapping. The selected proteins were further analyzed by matrix - assisted laser resorption/ ionization two - stage time of flight ( MALDI TOF/TOF) tandem mass spectrometry. The mass spectrometric data were re- trieved through the general proteomics standard (GPS)in Mascot database. Results Thirteen serum proteins were up - regulated by acupuncture, while two were down - regulated. Three proteins were selected for retrieval. Serum amyloid P was down - regulated while serine protease inhibitor and albumin was up - regulated by acupuncture. Conclusion Acupuncture protects the brain from ischemic injury by up - regualting the expression of serine protease inhibitor and promoting the albumin synthesis in the liver
