摘  要:【目的】探讨辛伐他汀对在高糖诱导下体外培养的人腹膜间皮细胞转化细胞因子-β1(TGF-β1)和结缔组织生长因子(CTGF)表达的影响。【方法】胰蛋白酶消化法从人腹膜组织中分离间皮细胞进行原代培养并传代;采用半定量反转录多聚酶链反应法(Semi—QuantitativeRT—PCR)检测细胞内TGF-β1和CTGFmRNA表达;酶联免疫双抗夹心法(ELISA)检测细胞上清液中TGF—β1和CTGF蛋白质水平,细胞沉淀用BCA蛋白检测方法测定细胞蛋白质含量,用以校正ELISA结果。【结果】辛伐他汀能显著降低高糖所致的腹膜间皮细胞TGF-β1和CTGF蛋白质水平(TGF—β1,P〈0.05;CTGF,P〈0.01),并下调其mRNA表达(TGF-β1,P〈0.01;CTGF,P〈0.05),两者在蛋白质和基因水平均呈量效关系。【结论】辛伐他汀能抑制高糖诱导下腹膜间皮细胞致纤维化细胞生长因子TGF-β1和CTGF的过度表达。[Objective]To investigate the effects of simvastatin on TGF-β1 and CTGF expressions in human peritoneal mesothelial cells (HPMCs) cultured in high glucose conditions. [Methods] HPMCs were isolated from human omenta by trypsin digestion method and subcultured. Semi-quantitative RT PCR was used to detect the mRNA expressions of TGF β1 and CTGF in HPMCs. Protein levels of TGF-β1 and CTGF in culture supernatants were measured by ELISA. Cells protein concentration measured by trace BCA method was to cor rect the ELISA assay results. [Results] Simvastatin significantly decreased high glucose-induced TGF-β1 and CTGF expressions in a dose dependent manner both in protein and gene levels ( P 〈0.05). [Conclusion] Simvastatin can inhibit expressions of TGF-β1 and CTGF in HPMCs cultured under high glucose conditions.
