腹腔注射乳化异氟醚对大鼠气道上皮组织β防御素mRNA表达的影响

摘  要:目的本研究旨在探讨腹腔注射乳化异氟醚对大鼠气道上皮细胞中β防御素BD-1、BD-2以及促炎症因子TNF-α、IL-1β、IL-8 mRNA表达水平的影响。方法健康清洁级SD大鼠36只随机分成6组,分别为A^F组。A组经腹腔注射生理盐水,B组经腹腔注射30%脂肪乳,C^F组分别经腹腔注射0.25、0.5、1.0、1.5倍ED50量8%乳化异氟醚。各组均于注药后1 h处死大鼠,取其气管黏膜组织。从每个组收获的组织中检测β防御素BD-1、BD-2和促炎症因子TNF-α、IL-1β、IL-8的mRNA表达水平。结果在腹腔注射乳化异氟醚的大鼠气管上皮组织中除BD-1和IL-8 mRNA无变化外,BD-2和促炎症因子TNF-α,IL-1βmRNA表达水平均增加,且随乳化异氟醚用量的增加而增强。结论本实验说明腹腔注射乳化异氟醚能在转录水平上调节大鼠气道上皮组织β防御素及一些促炎症因子mRNA的表达水平,且乳化异氟醚以剂量依赖的方式显著增加大鼠气道上皮细胞的β防御素BD-2以及这些促炎症因子mRNA的表达水平。Objective To examine the expression of β defensin genes and inflammatory cytokines genes in rat airway epithelium after intraperitoneal injection of emulsified isoflurane.Methods Thirty-six SD rats were assigned into six groups(A-F) randomly.The tissue from rat airway epithelium were harvested at 1 h after intraperitoneal injection of normal saline,30% fat milk and 8% emulsified isofluranes of 4 different dosages(0.25,0.5,1.0 and 1.5 times of ED50).Then expressions of β defensin(BD-1,BD-2),tumor necrosis factor-alpha(TNF-α),interleukin beta(IL-1β),IL-8 mRNA were measured by semiquantitative polymerase chain reaction(PCR) using GAPDH as an internal standard.Results Except BD-1 and IL-8 mRNA,marked increases in expressions of TNF-α,IL-1β and BD-2 genes were observed with a dosage-dependent manner.Conclusion The expressions of BD-2 genes and inflammatory cytokines genes(IL-1β,TNF-α)in rat airway epithelium increase significantly in a dosage-dependent manner after intraperitoneal injection of emulsified isoflurane.
