益气化瘀方对大鼠腰神经根损伤后纤维连接蛋白的作用

摘　　要：目的:探讨益气化瘀方对神经再生修复过程中神经肌肉接头部纤维连接蛋白(fibronectin FN)的作用。方法:大鼠L5神经根受压后,分别予以益气化瘀方灌胃治疗10、30和60d后取比目鱼肌。以FN作为细胞外基质标记,α-bunga-rotoxin(α-BTX)萤光结合剂显示乙酰胆碱受体(acetylchline receptor,AChR),采用免疫组织化学方法结合激光共聚焦扫描显微(Confocal Iaser Scanning Microscopy system)技术,观察大鼠比目鱼肌神经肌肉接头部FN的表达,NI H图像分析技术定量测定FN与AChR的重叠面积。结果:大鼠L5神经根损伤后10d和30d,益气化瘀方组FN的表达及与乙酰胆碱受体(Acetylchline Receptor,AChR)的重叠面积均明显高于模型组,60d时,益气化瘀方组FN的表达较模型组显著回落,AChR的形态再构建明显优于模型组。结论:FN的表达与运动神经终板的神经再支配状况密切相关,益气化瘀方可促进FN表达,加快神经再生修复进程。Objective：To study the effect of Yiqi Huayu Recipe on fibronectin（FN）in neuromuscular junctions during nerve regeneration.Methods：After L5 nerve root was compressed in rats,Yiqi Huayu Recipe was intragastically administered for 10 days,30 days,and 60 days respectively,and then soleus muscles were taken.The expression of FN in neuromuscular junctions of soleus muscle was observed using immunohistochemistry and confocal laser scanning microscopy techniques with FN as extracellular matrix marker and fluorescein-conjugatedα-bungarotoxin（α-BTX）to mark acetylcholine receptor（AChR）.The overlap areas of FN /AChR site was measured with NIH image technique.Results：Ten days and 30 days after L5 nerve root was compressed,the expression of FN and the overlap area of FN with AChR site in the Yiqi Huayu Recipe-treated group were markedly elevated than those in the untreated group.But at the 60th day,the expression of FN in the Yiqi Huayu Recipe group was obviously lower than untreated group and the morphology of neuromuscular junctions＇ reformation was better than untreated group.Conclusion：The FN expression is connected with motor endplate reestablishment after degeneration.Yiqi Huayu recipe may shorten this regeneration process.
