内障丸对青春期PCOS大鼠卵巢形态学和血清激素的影响

摘　　要：目的:探讨内障丸对青春期PCOS大鼠卵巢形态学和血清激素的影响。方法:以23日龄幼年SD清洁雌性大鼠为研究对象,应用皮下埋植左炔诺孕酮硅胶棒联合皮下注射HCG法建立青春期PCOS动物模型。运用免疫组化及光、电镜技术等方法,检测大鼠造模前后及给药前后卵巢形态学和血清激素的变化。结果:内障丸高、中、低剂量组大鼠用药后体重均明显低于模型组。内障丸治疗后卵巢色泽红润,高剂量组卵巢重量较模型组明显降低。36日模型组卵泡比例较对照组明显增加(P<0.01),颗粒细胞层厚度较对照组明显变薄(P<0.01),泡膜细胞层厚度较对照组明显增厚(P<0.05)。内障丸治疗后高、中、低剂量组囊状卵泡比例均明显降低(P<0.01),颗粒细胞层数均较模型组明显增加(P<0.05),其中低剂量组黄体组织数量增多明显(P<0.05)。造模后模型组E2含量变化不明显,而T含量显著升高(P<0.01)。内障丸治疗后高、中剂量组T含量均明显低于模型组(P<0.01)。结论:①皮埋左旋18-甲基炔诺酮联合HCC诱导SD幼年雌性大鼠PCOS动物模型卵巢病理改变及血内分泌改变与PCOS病人相似。②内障丸可以改善多囊卵巢大鼠卵巢形态学的异常,其促排卵的机制可能是通过对颗粒细胞和卵泡膜细胞的作用,降低多囊卵巢大鼠雄激素水平,从而促使卵巢排卵。③内障丸明显改善了多囊卵巢大鼠的高雄激素血症,对多囊卵巢大鼠异常的垂体及卵巢局部内分泌环境起到调节作用。Objective：The aim of this study was to investigate the effects of Neizhang Pill in puberty PCO rats＇ Ovarian morphology and Serumhormone.Method：Take the 23-day-old immature female SD rats as the object of study,progesterone-synchronized im-mature rats were used as the animal model for PCO by injectinghCG.Compared with normal control group,investigate the changement of the model group＇s Ovarian morphology and Serumhormone.Results：The body weight of the treatment groups significantly lightened than the control group.In PCO rats the number of Iuteum decreased but the number of cystic follicles increased contrarily.The cellular layer of ovary granulosa cell of the rat with PCO got thinner and thecal cell became thicker.BSHXT had the effect of increasing the cellular layer of ovary granulosa cell and decreasing the cellular layer of thecal cell for the rats with PCO.In PCO rats,serum T levels werehigher and in the treatment groups serum T levels significantly lower than the control group.Conclusions：①This study set up PCO rat model,the changement of ovarian pathology and incretion are similar as the patient of PCOS.②Neizhang Pill could improve internal secretion of pituitary and ovary of PCO rats through hyperinsulin.③Neizhang Pill could effect on PCO rats＇ovarian morphology and by this way to improve ovarian function.
