电针对吗啡戒断大鼠胸腺细胞PKA表达及胞内Ca^(2+)含量的影响

摘　　要：目的:探讨电针抗吗啡戒断大鼠胸腺细胞凋亡的作用机制。方法:将40只SD大鼠随机分为正常组,对照组,戒断组,电针组,共4组。连续20天递增量肌肉注射吗啡造成吗啡成瘾大鼠模型。电针组大鼠电针双侧足三里穴,每日1次,每次30min,治疗7天。采用激光扫描共聚焦显微技术,观察各组大鼠胸腺细胞胞内Ca2+的含量;运用免疫组化法检测PKA蛋白表达,观察各组大鼠胸腺细胞PKA蛋白表达。结果:与正常组相比,造模后对照组及戒断组胸腺细胞[Ca2+]i含量及PKA蛋白表达呈上升趋势;电针治疗后胸腺细胞[Ca2+]i含量及PKA蛋白表达则呈下降趋势。结论:电针具有调节吗啡戒断大鼠胸腺细胞[Ca2+]i含量及PKA蛋白表达的作用,这可能是电针抗吗啡戒断大鼠胸腺细胞凋亡的作用机制之一。Objective：To explore the mechanism of electroacupuncture （EA） on thymocyte apoptosis in morphine withdrawal rats. Methods：Forty SD rats were randomly divided into the normal group,the control group,the dependence group and the electroacupuncture group （EA group）. Rat abstinence syndrome model was established by continuous muscular injection of morphine to the hind limds for 20 days. EA therapy was given to rats in the EA group,EA was applied to ＂Zusanli＂ （ST36） for 30 min,once every day,for 7 successive days. Laser scanning confocal microtechnic was applied to observe the content of thymus intracellula Ca2＋ in every group rats. Immunohistochemical method was applied to observe expression of PKA in every group rats. Results：Compared with normal,the content of thymus intra-cellula Ca2＋ and expression of PKA in the control group and dependence group taked on increased tendency obviously. The content of thymus intra-cellula Ca2＋ and expression of PKA taked on reduced tendency obviously after EA therapy. Conclusion：EA possesses the effect of regulating the content of thymus intra-cellula Ca2＋ and expression of PKA,which might be one of the mechanisms of EA resisting thymus apoptosis in the morphine withdrawal rats.
