甘麦大枣汤、四逆散及四君子汤对慢性应激大鼠内源性神经生长因子水平的影响

摘  要:目的探讨疏肝、养心、健脾疗法对慢性应激大鼠内源性神经生长因子(NGF)水平的影响。方法120只SD大鼠分为正常组、模型Ⅰ组、模型Ⅱ组、甘麦大枣组、四逆散组和四君子汤组,每组20只。采用长期高压恒流电刺激足底法制作慢性应激动物模型。运用自发活动和穿越实验以及悬尾实验观察大鼠行为学变化。运用放射免疫分析法测定大鼠垂体、下丘脑和血浆中NGF水平。结果①与正常组[(1052.64±124.17)mm]比较,两模型组[(809.07±120.39)mm,(809.53±167.77)mm]和四君子汤组[(912.96±114.71)mm]大鼠的自发活动路程明显减少(P〈0.01);两个中药组[(995.17±142.8)mm,(1092.58±272.62)mm]明显多于两模型组(P〈0.01);②两模型组大鼠的穿越时间[(8.73±3.77)s,(8.07±3.16)s]明显减少(P〈0.01);与两模型组比较,三个中药组穿越时间[(13.3±6.15)S,(12.19±5.11)s,(11.7±4.75)S]明显多于两模型组(P〈0.01);(3)两模型组大鼠的站立次数[(4.79_-4-3.47)次,(5.30±3.76)次]明显减少(P〈0.01);三个中药组[(7.55±6.56)次,(8.12±4.99)次,(9.51±6.02)次]明显多于两模型组(P〈0.05);④两模型组大鼠的悬尾不动时间[(83.42±29.18)S,(85.89±31.10)s]明显增多(P〈0.01);与两模型组比较,三个中药组[(117.24±39.46)S,(121.87±39.88)S,(112.21±32.62)S]明显少于两模型组(尸〈0.01);⑤垂体中,甘麦大枣汤、四逆散以及四君子汤组NGF水平[(115.68±9.64)pg/mg,(209.63±17.44)pg/mg,(171.86±14.32)pg/mg]明显增加(P〈0.01);下丘脑,四君子汤组[(12.02±11.43)pg/mg]与正常和模型Ⅱ组[(40.61±39.7)pg/mg,(39.46±31.88)pg/mg]相比NGF水平明显降低(P〈0.05)Objective To investigate the effects of Ganmaidazao Decoction(GMDZD) ,Sinisan(SNS) and Sijunzi Decoction (SJZD) recipe on endogenous nerve growth factor (NGF) of rat with chronic stress. Methods In order to establish the rat models with chronic stress, the feet of animals were stimulated with a pat- tern of electrical impulses (long term,high voltage and constant current). The alterations of behavior of animals were observed and quantified by open-field and tail-suspension test. l^vels of NGF in pituitary, hypotholamus and blood plasma were quantified using radio-immunoassay (RIA) method. Results 1 ) In comparision with the wild-type (WT) control rats, locomotor activity decreased significantly in the models and groups treated by Chinese medicinal herbs (P〈 O. 01 ). In addition, the Chinese medicinal herbs treated groups exhibited increased locomotor activity than the model groups (P 〈 0.01 ). 2) The cross time of the model groups decreased significantly compared with the WT controls (P 〈 0.01 ). Meanwhile,three Chinese medicinal herbs treated groups displayed longer cross time than the model groups (P〈 0.01 ). 3 )The vertical movement times of the model groups decreased significantly compared with the WT controls (P 〈 0.01 ). Meanwhile, three Chinese medicinal herbs treated groups displayed in- creased vertical movement times than the model groups (P 〈 0.05). 4) In the tail-suspension test, the duration of immobility increased significantly in the model groups compared with the WT controls (P 〈 0.01 ). However,the duration of immobility of three Chinese medicinal herbs treated groups were less than the model groups (P 〈0.01 ). 5) Compared to the model groups,the level of NGF in pituitary of animals treated by GMDZD and SNS in- creased significantly (P 〈 0.01 ). In the hypothalamus ,the level of NGF in SJZD treated groups decreased in com- parision with the WT control and model groups (P 〈 0.05 ). Whereas, GMDZD and
