摘  要:目的研究BAFF-R-IgG4mut融合蛋白对BXSB狼疮鼠B淋巴细胞的影响。方法体外观察BAFF-R-IgG4mut融合蛋白对BAFF刺激的B细胞活性的影响;100μl的PBS、200μg人IgG4和100μg BAFF-R-IgG4mut融合蛋白分别经腹腔注射10周龄雄性BXSB狼疮鼠,每周注射3次,共注射5周。分别检测狼疮鼠外周血BAFF水平、外周血和脾脏组织中B220+CD5-、CD3+CD4-、CD8+T和CD3+、CD4+、CD8-T细胞的表达以及狼疮鼠的存活率。结果BAFF-R-IgG4mut融合蛋白组外周血BAFF以及外周血、脾脏组织中B220+CD5-细胞较对照组下降(P<0.01),实验组狼疮鼠存活率较对照组延长(P<0.01)。结论BAFF-R-IgG4mut融合蛋白可抑制BXSB狼疮鼠B细胞活性。Objective To study the effect of BAFF-R-IgG4mut fusion protein on B lymphocytes of routine BXSB lupus. Methods BXSB male mice were randomly divided into three groups and treated with PBS, human IgG4 and BAFF-R-IgG4mut fusion protein three times a week for five weeks. Blood samples were collected from the tail veins to examine the lymphocyte status in peripheral blood and spleen. BAFF level in peripheral blood were detected with ELISA. Results BAFF-R-IgG4mut was able to completely inhibit BAFF-mediated co-stimulation of B cell proliferation in vitro. Treatment of BXSB lupus mice with BAFF-R-IgG4mut fusion protein for 5 weeks resulted in significant B-cell reduction both in the peripheral blood and in spleen. Compared with control, animals treated with BAFF-R-IgG4mut had 100% survival at 25 weeks of age compared with 0% survival in the control group (P〈0. 01). Conclusions These results indi- cate that BAFF-R-IgG4mut fusion protein can inhibit the activity of locking B-cell and may have a pathogenic effect on the development of renal disease.
