摘 要:目的观察抗组胺药苯海拉明对小鼠记忆的影响。方法将小鼠分为生理盐水组(NS组)和苯海拉明组(ND组),根据苯海拉明剂量的不同,ND组又分为ND1(2.0 mg/kg)、ND2(4.0 mg/kg)、ND3(6.0 mg/kg)组,每组10只。利用避暗实验观察小鼠潜伏期(latency)和错误次数(error tim es),评估药物对小鼠记忆功能的影响。结果在避暗实验中,用药后第1天,与NS组相比,ND1、ND2、ND3组潜伏期缩短(P<0.01),错误次数增多(P<0.01);与ND1组相比,ND3组潜伏期缩短,错误次数增多,差异有显著性(P<0.05);用药后第2、3天,各组错误次数、潜伏期的差异均无显著性(P>0.05)。结论苯海拉明能损害小鼠记忆功能,且呈剂量依赖性,记忆损害在停药1天后迅速消失,提示不会造成持久的记忆障碍。Objective To investigate the effects of an antihistamine,diphenhydramine on the memory of mice.Methods The mice were divided into normal saline group(NS group) and diphenhydramine groups(ND groups),which were subdivided into ND1(2.0 mg/kg),ND2(4.0 mg/kg) and ND3(6.0 mg/kg) groups(n=10 each) according to the doses of diphenhydramine.The latency and error times in the step-through passive avoidance task were recorded to evaluate the effects of diphenhydramine on the memory of mice.Results In the step-through test,on the first day,as compared to NS group,the latency decreased(P〈0.01),and the error times increased(P〈0.01) in ND1,ND2 and ND3 groups;as compared to ND1 group the latency decreased and the error times increased in ND3 group,with significant difference(P〈0.05).There was no significant difference in the latency and error times between each group on the second day and third day(P〉0.05).Conclusion Diphenhydramine can impair the memory function of mice in a dose-dependent manner.The memory impairment disappears quickly within the 1st day of drug withdrawal,implying that diphenhydramine causes no persistent dysfunction in memory.
