摘  要:以蝴蝶兰无菌种子为材料,对蝴蝶兰种子消毒和萌发、原球茎诱导、增殖和分化培养、芽生根培养基、壮苗培养基配方进行研究。结果表明,种子经次氯酸钠和洗衣粉消毒,有利于促进种子萌发和原球茎的形成;最适合种子萌发的培养基为MS+香蕉泥100g/L+马铃薯50g/L+NAA 0.5mg/L;原球茎的形成及增殖培养以MS+6-BA 2.0mg/L+NAA0.1mg/L+香蕉泥100g/L为佳,分化培养基为MS+6-BA 0.1mg/L+NAA 0.1mg/L+香蕉泥80g/L+蔗糖20g/L+活性炭2.0g/L+卡拉胶7.0g/L。小苗生根培养基为MS+NAA 0.1mg/L+香蕉泥100g/L+蔗糖20g/L+卡拉胶7.0g/L+活性炭2.0g/L,生根率达98%以上;壮苗培养基为MS+香蕉泥80g/L+马铃薯汁50g/L+蔗糖20g/L+卡拉胶7.0g/L+活性炭2.0g/L。经过适当驯化处理,将壮苗移栽到疏松的水草或苔藓基质中,注意温度、湿度及光照管理,成活率可达90%以上。An experiment was conducted to study the effect of sterilization methods and culture media on germination of seeds.Various induction,multiplication,differentiation and culture media were evaluated for protocorm multiplication,bud rooting and vigorous plantlet growth using sterile seed of Phalaenopsis.The results showed that seeds sterilized with detergent and 10% NaClO were best suitable for the seed germination and protocorm formation.The suitable medium for seed germination was MS+ banana mud 100g / L+ potato 50g / L+NAA 0.5mg / L.The best culture medium for the formation and multiplication of protocorm was MS+6-BA 2.0mg / L+NAA 0.1mg / L+ banana mud 100g / L,and the differentiation medium was MS+6-BA 0.1 mg / L+NAA 0.1 mg / L +banana mud 80g / L+sugar 20g / L +active carbon 2.0g / L +carrageenan 7.0 g / L.The rooting medium for young plantlet was MS +NAA 0.1mg / L +banana bud 100g / L +sugar 20g / L + carrageenan 7.0g / L + active carbon 2.0g / L which resulted in more than 98% rooting rate.For culturing vigorous plantlet,MS +banana mud 80 g / L +potato 50 g / L +sugar 20 g / L + carrageenan 7.0 g / L + active carbon 2.0 g / L was found to be the best medium.The vigorous plantlets,obtained using our methods were transplanted into loose waterweeds or moss with suitable management of temperature,humidity and light resulted in more than 90% survival rates.
