摘 要:目的探索金属硫蛋白和锌对急性贫铀中毒的解毒效果。方法给予Wistar大鼠贫铀溶液造成急性贫铀中毒的动物模型,利用ZnSO4和金属硫蛋白(metallothionein,MT)解毒。Wistar大鼠70只,随机分为7组,每组10只:①正常组(N);②贫铀组(DU);③ZnSO4预防组(Zn-P);④MT预防组(MT-P);⑤ZnSO4治疗组(Zn-T);⑥MT治疗组(MT-T);⑦ZnSO4预防+MT治疗组(Zn+MT)。测定血、尿和肾脏标本的铀含量。观察内/外源性的MT作用对大鼠急性贫铀中毒的解毒效果,同时观察不同解毒措施对血常规和肝、肾功能的影响以及肾脏病理学变化。结果Zn预防组、MT预防组和MT治疗组均有明显的解毒效果[肾脏和尿液中贫铀含量明显增高(P<0.05)],其中又以Zn预防组的解毒效果最好,病理变化较轻。同时发现在使用MT或Zn进行干预的各个干预组中,没有表现出明显的血常规和肝、肾功能指标的改变。结论预防性给予ZnSO4诱导产生内源性金属硫蛋白,能更好发挥对贫铀中毒的解毒效果。Objective To investigate the effect of the metallothionein (MT) and zinc, as chelating antidotes on depleted uranium acute poisoning. Methods Totally 70 healthy adult Wistar rats were divided into 7 groups: ① Normal group (N) ; ②Depleted uranium (DU) ; ③ ZnSQ prevention group (Zn-P) ; ④ MT prevention group (MT-P) ; ⑤ ZnSQ treatment group (Zn-T) ; ⑥ MT treatment group (MT-T) ; ⑦ ZnSO4 prevention + MT treatment group ( Zn + MT). Rat model of acute depleted uranium poisoning was induced by a depleted uranium solution. In prevention group, ZnSO4 solution at 20 mg · kg^- 1 · d^ - 1 or MT solution at 5 mg · kg^- 1 · d^ - 1 was injected intraperitoneally to correponding rats for 2 d, and then those rats were established to model of model of acute depleted uranium poisoning. While for the rats of treatment groups, ZnSO4 solution or MT solution at above doses were given to depleted uranium acute poisoning rats for 3 d. All the animals were sacrificed to take blood, kidney and liver samples on the 5th day after injection of DU. The effects of detoxication of MT and Zn to DU were observed by testing blood routine, liver and kidney function and renal pathological changes. Results Different intervention measures had a certain detoxifying effect, especially ZnSO4-induced generation of MT in vivo, resulting in some slight pathological changes in kidney. No significant change was observed in parameters of blood routine tests and renal function in different intervention groups. Conclusion MT plays a role in detoxification of acute depleted uranium poisoning with no obvious side effects on liver and kidney.
