摘 要:目的利用胶体金技术制备明胶胶体金探针并用胶体金标记明胶-阿拉伯胶空微囊,为下一步利用胶体金技术研究药物在体内的分布探索新的试验方法。方法利用胶体金表面带负电荷而壳聚糖、明胶等微囊材料表面带正电荷,它们之间可形成非共价键的静电吸引而牢固结合的原理,以明胶为微囊囊材,使之先与胶体金特异结合,再用标记过的明胶以复凝聚包囊工艺制备明胶-阿拉伯胶微囊。最后利用电镜观察胶体金在体内的分布,得到胶体金标记微囊在动物体内的分布情况。结果胶体金标记好的明胶-阿拉伯胶空微囊被成功制备。结论胶体金能很好地标记明胶微囊类药物。Objective To prepare glutin colloidal gold probes by the colloidal gold technique and to label glutin-arabic gum empty microencapsules by colloidal gold for searching the new experimental method to further study the drug distribution in vivo with this technique. Methods To use the principle in which colloidal gold possessing negative charge in its surface and glutin possessing positive charge in its surface can form electrostatic attraction with non- covalence and combine firmly. Firstly, glutin as the microencapsule material was used to be specially combined with colloidal gold, the glutinarabic gum microencapsules were prepared by labeled glutin and the complex coacervation microencapsulated technology. Then, electron microscope was used to observe the colloidal gold distribution in vivo for obtaining the distribution status in animal body of microencapsules labeled by colloidal gold. Results The glutin- arabic gum empty microencapsules labeled by colloidal gold were successfully prepared. Conclusion Glutin microencapsule drugs can be labeled well by colloidal gold.
