摘  要:目的观察芪丹颗粒对博莱霉素A5诱导的肺纤维化大鼠病变严重程度及FasL、Caspase-3表达的影响,以探讨其可能的作用机制。方法70只健康雄性清洁级Sprague-Daw ley(SD)大鼠,随机分成4组。28、42d正常对照组(对照组)各5只,模型组各10只,芪丹组各10只,氢可组各10只。对照组气管内和灌胃均用生理盐水,余SD大鼠气管内灌注博莱霉素A5诱导肺纤维化,第15d开始每日分别给予芪丹颗粒剂灌胃(芪丹组3125mg/kg)、腹腔注射氢可(氢可组25mg/kg)。分别在第28、42d处死。苏木素-伊红(HE)染色观察肺泡炎及纤维化程度,免疫组化观察石蜡切片中FasL及Caspase-3的表达。结果芪丹颗粒剂组较氢可组及模型组肺纤维化及肺泡炎程度轻。FasL表达芪丹组28、42d与对照组差异有统计学意义,P<0.01;与模型组及氢可组比较差异无显著性。Caspase-3芪丹组与模型组及氢可组比较表达差异无显著性意义。结论芪丹颗粒剂可减轻博莱霉素A5诱导的肺纤维化大鼠的肺纤维化及肺泡炎。肺纤维化形成后期应用芪丹颗粒剂对FasL及Caspase-3的表达无减轻。Objective To investigate the influence of Qi-dan granule on the severity of leision and the expression of FasL and caspase-3 in bleomycin As-induced pulmonary fibrosis in rats,in order to explore its probable mechanisms. Method A total of 70 health male Sprague-Dawley(SD) rats of clean grade were randomly divided into 4 groups : normal group ( 10 rats ; 28-, 42-days subgroups ) , BLM group ( 20 rats ; 28-, 42-days subgroups), Qi-dan granule group ( 20 rats ; 28-, 42-days subgroups ), cortisol group ( 20 rats ; 28-, 42-days subgroups). Saline was used in intratracheal instillation and gastric perfusion in control group, while bleomyein A5 was used in other groups to induce the pulmonary fibrosis. Since the 15th day, gastric perfusion with Qi-dan granule and intraperitoneal injection with cortisol were undergone every clay. Animals were killed at the 28th and 42nd days respectively. Microscopy was performed with HE staining to recognize the severity of alveolitis or the degree of pulmonary fibrosis, lmmunohistochemical staining was performed to recognize the expression of FasL and caspase-3. Results Qi-dan granule decreased the severity of inflammatory cell as well as the degree of fibrosis. The expression of Fasl, in Qi-dan granule group(28-,42-days subgroups) was significantly different from that in control group( P 〈0. 01 ) ,whilethe expression of FasL and caspase-3 were similar to that in cortisol and model groups ( all P 〉 0. 05 ). Conclusions Qi-dan granule could reduce the degree of alveolitis and fibrosis in rats with pulmonary fibrosis induced by bleomycin As,but it could not influence the expression of FasL and caspase-3 at the latter stage of pulmonary fibrosis.
