摘 要:目的观察大鼠脑挫伤后不同时间段低氧诱导因子HIF-1α的表达变化规律,探讨其在脑挫伤过程中的作用机制及法医学实践中的应用。方法参照Feeney法建立大鼠闭合性脑挫伤模型,将实验动物分为对照组和挫伤后1h,4h,12h,48h,72h,7d,14d共8组,运用Western blot方法检测HIF-1α蛋白的表达。结果伤后1h可观察到少量HIF-1α表达,4h后表达开始增加,48h达高峰,到14d时与对照组无明显差异。结论脑挫伤后HIF-1α的表达呈现出一定的时序性规律,有望应用于脑挫伤经过时间的法医学鉴定。Objective To investigate the changes of hypoxia-inducible factor( HIF) -1α expression in the different posttraumatic intervals after cerebral contusion in rats,and to explore its mechanism and the forensic significance. Methods Based on Feeney' s method, the rat model of cerebral contusion was established. At 1 h,4 h, 12 h,48 h,72 h,7 d, 14 d after cerebral contusion, the expression of HIF-1α protein was detected by Western blot. Eight normal Wistar rats were chosen as controls. Results The expression of HIF-1α protein was observed at 1 h after injury, increased at 4 h, peaked at 48 h, and decreased to the level of the controls at 14 d. Conclusion The sequential expression of HIF-1 ot may be used for the estimation of posttraumatic intervals after cerebral contusion in forensic practice.
