摘 要:目的考察草豆蔻、草果、白豆蔻挥发油对模型药士的宁经皮渗透的影响。方法采用改良Franz扩散池法,以小鼠皮肤为透皮屏障,用HPLC法测定士的宁的含量,并以氮酮为对照,考察不同浓度的草豆蔻、草果及白豆蔻油对士的宁的促渗效果。结果在同系列不同浓度中,3%氮酮、7%草豆蔻油、5%草果油及5%白豆蔻油对士的宁的促透效果最好,对9mg·mL-1士的宁的12h平均累积渗透量分别为1.577、1.949、1.716、1.359mg·cm-2·h-1,其增渗倍数分别为2.719、3.284、3.312、2.334倍。经t检验,5%草果油、7%草豆蔻油稳态渗透速率(Jss)明显高于阴性对照组(P<0.05)。结论7%草豆蔻油、5%草果油均能明显促进士的宁的经皮渗透。OBJECTIVE To investigate the effects of volatile oils of Alpinia katsumadai Hayata (AKH) ,Amomum tsao -ko (ATK) and Amomum kravanh Pierre(AKP) on percutaneous penetration of Strychnine through mouse skin in vitro. METHODS By using an improved Franz diffusion cells, the enhancing effects of 5 different concentrations of these 3 kinds of volatile oils on the percutaneous penetration of Strychnine were observed and compared with that of Azone. The content of Strychnine was determined by HPLC. RESULTS The permeability - promoting effects of 3% of Azone,7% volatile oil of AKH,5% of AT - K and 5% of volatile oil of AKP on strychnine were the best and the 12 h average cumulative permeation amount of strychnine were 1. 577,1. 949,1. 716, 1. 359 mg·cm^-2·h^-1, respectively. The enhancement ratios were 2. 719,3. 284,3.312 and 2. 334, respectively. By t - test, the permeability coefficients of 7% volatile oil of AKH and 5% AT- K were significantly higher than that of the negative control group( P 〈 0.05 ). CONCLUSION 7% volatile oil of AKH and 5% volatile oil of AT - K could enhance permeation of Strychnine in vitro obviously.
