摘 要:目的研究天然酮类化合物抗柯萨奇病毒B5(CVB5)的作用及其机理。方法通过改变给药途径,观察病毒引起的细胞病变效应(CPE),MTT法检测细胞活性,评价酮化合物在Hep-2细胞培养中的抗CVB5作用。结果酮具有抗CVB5的活性,对Hep-2细胞的TC50为54.44μg·mL-1,其EC50分别为26.378、25.813、25.405μg·mL-1,治疗指数(TI)分别为2.064、2.109、2.143;在32μg·mL-1时,对CVB5的抑制率达50%。结论酮化合物通过直接杀灭病毒、阻止病毒的吸附、抑制病毒的生物合成而发挥其抗CVB5的作用;但需对结构做进一步的修饰,以降低其毒性。OBJECTIVE To study the anti - Coxackievirus B5 ( CVB5 ) effect and mechanism of natural xanthone in vitro. METHODS By changing the drug adding occasion, the virus - induced cytopathic effect (CPE) was observed and MTT colorimetric assay was used to evaluate its antiviral effect. RESULTS Xanthone showed significant anti - CVB5 activity. Its 50% toxic concentration on Hep -2 cell was 54. 44 μg·mL^-1. The 50% inhibitory concentrations were 26.378, 25. 813 and 25. 405μg·mL^-1 ,respectively. The treatment indexs were relatively figured out as 2. 064, 2. 109 and 2. 143. Its anti - CVB5 inhibitory rate was more than 50% at the dose of 32μg·mL^-1. CONCLUSION The natural xanthone exerts its anti - CVB5 activity by direct killing,anti - virus adsorption, anti - virus biosynthesis, yet further structure modification is needed due to its toxic effects to cells.
