摘  要:目的采用HPLC法同时测定槐角中芦丁和槐角苷的含量。方法采用Diamonsil C18柱(150mm×4.6mm,5μm),流动相为甲醇-1%冰醋酸-乙腈(38∶57∶5),流速1.0mL·min-1,检测波长为260nm。结果杂质峰出峰时间快,芦丁与槐角苷的出峰时间适中,分离度好;两者0.125~2.000μg·mL-1与峰面积呈良好的线性关系。芦丁、槐角的苷平均回收率分别为98.18%、97.33%,RSD分别为1.48%、1.92%(n=6)。结论所建方法适用于槐角中芦丁和槐角苷含量的同时测定。OBJECTIVE To develop RP - HPLC for the determination of rutin and sophoricoside in Fructus sophorae simultaneously. METHODS The extracts of Fructus sophorae were separated on a Diamonsil C18 column ( 150 mm×4.6mm,5μm). Methanol - 1% glacial acetic acid- acetonitrile (38 : 57 : 5 )was used as mobile phase. The flow rate was 1.0 mL·min^-1. The effluents were monitored at 260 nm. RESULTS The peaks of impurity were eluted rapidly and the peaks of rutin and sophorieoside were separated well with a proper retention time. The linear ranges of rutin and sophoricoside were 0.125 -2. 000 μg·mL^-1. The average recovery rate for rutin was 98.18% with RSD of 1.48% (n = 6)and that for sophorieoside was 97.73% with RSD of 1.92% (n = 6 ). CONCLUSION This method is suitable for the determinination of rutin and sophorieoside in fruetus sophorae simultaneously.
