摘  要:目的探讨中药加味玉屏风颗粒对变应性接触性皮炎(CHS)小鼠模型的治疗作用及对TNF-α表达的影响。方法用DNFB诱导建立小鼠CHS动物模型,分为生理盐水组,氢化可的松组及中药低中高剂量组(国家药典人体用量的10倍、20倍、40倍),测量给药后的小鼠耳片厚度差、重量差、局部炎细胞浸润以及血清中TNF-α的改变。结果加味玉屏风颗粒组小鼠耳厚度(0.1387±0.0183、0.1116±0.0243、0.0913±0.0173)、重量(5.802±0.9610、3.747±1.2831、2.956±1.1214)的改变均小于生理盐水组(P<0.05),局部单一核细胞及多核细胞计数明显减少,血清TNF-α(154.9217±21.6511、137.4770±15.0718、104.5124±16.6126)亦低于对照组(204.5643±35.7699)(P<0.05)。结论加味玉屏风颗粒可有效抑制CHS小鼠皮肤炎症,其作用机制与其下调TNF-α相关。Objective investigate the therapeutic effect of modified yupingfeng granule on CHS mice and the influence on expression of TNF-α. Methods Establish DNFB sensitized CHS mouse model, and divided into NS group, positive control group, low-dosage, moderate-dosage and high-dosage groups (10 times, 20 times and 40 times of what is regulated for human administration in the State Pharmacopeia. ), observe the changes of clinical indexes of the mice after administrated of the granules, Then the expressions of TNF-α in the mouse peripheral blood were detected by ELISA. Results The changes of thickness(0. 1387 ±0.0183,0. 1116 ± 0.0243,0. 0913 ± 0. 0173 ) ,weights of the mice auricle(5. 802 ± 0. 9610,3. 747 ± 1,2831,2. 956 ± 1. 1214 ) were significant decreased respectively with the NS group ( P 〈 0.05 ). the local infiltration of mononuclear and polynuclear cells were reduced, the level of TNF-α in experiment group ( 154. 9217 ± 21. 6511, 137.4770 ± 15. 0718,104. 5124± 16. 6126) was significantly lower than the NS group (204. 5643 ± 35. 7699) ( P 〈 0.05 ). Conclusions Modified yupingfeng granule can effectively inhibit allergic contact dermatitis in mice. The regulating of the expression of TNF-α maybe one of its mechanisms
