摘 要:目的研究系统性红斑狼疮(SEE)患者外周血白细胞中IKB激酶(IKK-α)、干扰素(IFN)-αmRNA的表达,并检测血浆中IFN-α的水平,以探讨SLE患者中IKK—α在IFN—α产生中的作用。方法SYB Rgreen dye I实时定量聚合酶链反应(PCR)方法检测外周血白细胞IKK-α和IFN-α的表达;酶联免疫吸附试验(ELISA)法检测血清IFN吨的水平。结果①SLE患者外周血IKK-αmRNA表达高于对照组(P〈0.05);在活动组SLE患者中IKK—αmRNA的表达高于非活动组SLE患者(P〈0.01)。②SLE患者IFN-αmRNA的表达低于对照组(P〈0.01),IFN-αmRNA的表达在非活动组SLE患者中低于活动组SLE患者(P〈0.01)。③SLE患者血清中IFN-α的水平高于对照组(P〈0.01),其中,活动组SLE患者血浆IFN-α水平显著高于非活动组患者(P〈0.05);SLE患者血浆中IFN-α浓度与抗双链DNA(dsDNA)抗体呈正相关(P=001),与补体C3水平呈负相关(P=0.005)。④SLE患者IKK-αmRNA的表达与血浆中IFN-α的水平呈正相关(P=0.001).结论SLE患者IKK-αmRNA的表达明显增高,且与血浆中IFN-α的水平呈正相关;而血浆中IFN—α的水平与SLE的发病及病情活动相关,提示IKK-α可能在SLE的发病中发挥重要的作用。Objective To investigate the mRNA expression of IKB kinase (IKK-α) and interferon-ix (IFN-α) in the peripheral blood leukocytes of patients with systemic lupus erythematosus (SLE), and to explore the role of IKK-α in the production of IFN-α in SLE patients. Methods SYBR green dye I based real-time quantitative PCR was used to detect the mRNA expression levels of IKK-α and IFN-α in the peripheral blood leucocytes of SLE patients and healthy controls. Serum levels of IFN-α were measured with ELISA method. Results IKK-αmRNA expression levels in SLE patients were significantly higher than those of normal controls (P〈0.05). IKK-α mRNA expression levels in SLE patients with active disease were significantly higher than patients with stable disease (P〈0.01). IFN-α mRNA expression level in SLE patients was significantly lower than that of the normal controls (P〈0.01). IFN-αmRNA expression levels in SLE patients with active disease were significantly higher than patients with stable disease (P〈0.01). Serum levels of IFN-α in SLE patients with active disease was significantly higher than that of the normal controls and patients with stable disease (P〈0.05). The anti-dsDNA antibody correlated positively, and complement C3 correlated negatively with serum concentration of IFN-α. IKK-αmRNA expression levels in SLE patients correlated positively with serum concentration of IFN-α. Conclusion IKK-α correlates positively with serum concentration of IFN-α. The IFN-α level is significantly correlated with disease activity. This suggests that IKK-α may play an important role in the pathogenesis of SLE.
