白细胞介素6受体基因多态性与代谢综合征相关性研究

摘　　要：目的探讨白细胞介素6受体(IL-6R)基因-183A/G(rs4845617A/G)和Asp358Ala(rs8192284 A/C)多态性与代谢综合征(MS)的相关性。方法 PCR-RFLP法分析IL-6R基因多态性与BMI、腰围、胰岛素敏感性、胰岛素分泌水平、MS的相关性。结果 rs4845617 A/G和rs8192284 A/C两位点间呈连锁不平衡;IL-6R基因多态性与北京市东城区常住汉族人群MS发病率、胰岛素分泌功能、腰围、BMI、胰岛素敏感性指数、TG水平相关。结论 IL-6R基因变异在MS发病中具有一定作用。Objective To examine the relationship between genetic variants in the IL6R gene and MS in the Han nationality. Methods Using the method of PCR-RFLP we identified 2 functional single nucleotide polymorphisms （SNPs） in the IL-6R gene： a Asp→Ala substitution at position 358（rs8192284 A/C） and a variant in the promoter-183 region （rs4845617 A/G）. The study population consisted 2456 subjects of the Han nationality from Dong-Cheng District of Beijing. Results The rs4845617A/G and rs8192284A/C were in linkage disequilibrium（D＇=0. 61 X^2 =52.8 P〈0. 05）. The A/A group （n=361）, A/C group （n=568）and C/C group （n=198） in rs8192284 SNP were studied and showed that rs8192284 A/C） polymorphism was associated with BMI, WC, Fins, 2hIns, ISI and prevalence of MS （all P% 0. 05）. The prevalence of MS in the 3 groups was 22.8%, 16.9%, 11.1%, respectively（all P = 0. 02）. Condusions IL-6R gene polymorphism is significantly associated with MS in Han nationality.
