传染性支气管炎病毒在鸡胚成纤维细胞中适应及复制的生物学特性研究

摘　　要：将传染性支气管郯病毒ＩＢＶ－ＮＨ、Ｔ、Ｈ５２、Ｍ４１适应鸡胚成纤维细胞（ＣＥＦ），连续传代获得毒价稳定细胞适应毒并对其生物学特性进行研究。观察了ＩＢＶ致ＣＥＦ细胞病变效应的特征，测定ＴＣＩＤ５０滴度为１０６．０～１０６．０。细胞培养病毒提纯后负染置电镜下进行形态学观察，见到典型的冠状病毒粒子。病毒经胰酶处理显示出血凝特性，该血凝性能被特异性血清所抑制。表明ＣＥＦ细胞病变（ＣＰＥ）确为ＩＢＶ所致。通过对４株ＩＢＶ在ＣＥＦ细胞中最佳培养时间及繁殖量关系的摸索，证实了３６～７２ｈ释放到培养液中有活性的病毒粒子达高峰，ＴＣＩＤ５０最高可达１０７．０／ｍｌ。The Infectious Bronchitis Viruses (IBV) were adapted and propageted in the material of chicken embryo fibroblasts (CEF).The stable TCID 50 titer of IBV NH、T、H 52 and M 41 strains was gained by serial passage.Studies on the biological feature were carried out by several assays.The characteristics of cytopathic effect (CPE) of IBV causing CEF cell were observed.The TCID 50 titers of several IBV were from 10 6.0 to 10 7.0 .The typical corona virus was sbserved with electro microscope by stample of purified IBV of CEF cell culture.The heamagglutinating activity of virus could be tested after trypsin handling and it could be inhibited by specific serum.The research results proved that CPE of CEF was caused by IBV.The titer of active viruses of cultural liquid was highest from 36 hours to 72 hours according to the relation between perfect selection of time for cultured IBV and capacity for propagation.The TCID 50 of the highest IBV strain was 10 7.0 /ml.
