摘  要:目的:建立高效液相色谱法同时测定妇科栓剂中氧化苦参碱、苦参碱和小檗碱3个有效成分的含量。方法:采用Kro-masil C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-乙腈-乙酸铵缓冲溶液(浓氨水调pH至8.5)为流动相进行梯度洗脱,流速1.0 mL.min-1,检测波长220 nm(氧化苦参碱、苦参碱)、265 nm(小檗碱),柱温为室温。结果:氧化苦参碱、苦参碱和小檗碱线性范围分别为0.095~2.37μg(r=0.9999),0.129~3.22μg(r=0.9999),0.362~9.05μg(r=0.9999);平均加样回收率(n=5)分别为100.2%,99.2%,98.9%。结论:本方法的精密度、稳定性、重复性和加样回收率均较好;所建立的HPLC方法准确、快速,可用于妇科栓剂中氧化苦参碱、苦参碱和小檗碱3个主要成分的含量测定。Objective:To develop an HPLC method for determination of three components(oxymatrine,matrine and berberine) in Fuke suppository.Methods:Analysis was performed on a Kromasil C18 column(250 mm×4.6 mm,5 μm) with the mixture of methanol,acetonitrile and ammonium acetate buffer solution(adjust pH to 8.5 with stronger ammonia water) as mobile phase in gradient elution,and the flow rate was 1.0 mL·min-1;The detection wavelength was 220 nm for oxymatrine and matrine and 265 nm for berberine;The column temperature was room temperature.Results:The calibration curves of oxymatrine,matrine and berberine were linear in the ranges of 0.095-2.37 μg(r=0.9999),0.129-3.22 μg(r=0.9999),0.362-9.05 μg(r=0.9999),respectively;The average recoveries(n=5) of the three components in Fuke suppository were 100.2%,99.2%,98.9%,respectively.Conclusion:The accuracy,stability,repeatability,and average recovery of the method are satisfying,and the three components(oxymatrine,matrine and berberine) of Fuke suppository can be rapidly and accurately quantified by HPLC.
